摘  要:为考察复方灵芝对小鼠的镇静、催眠作用,并研究其对脑内神经递质含量的影响。将小鼠随机分为空白对照组[蒸馏水,0.2 mL/(10 g·bw)]、阳性药物组[艾司唑仑,0.26 mg/(10 g·bw)]、复方灵芝组[126 mg/(10 g·bw),以生药量计],每组10只,小鼠每天灌胃给药1次,连续给药8 d。以小鼠入睡率、睡眠潜伏期和睡眠持续时间为药效指标,考察戊巴比妥钠协同下复方灵芝的镇静、催眠作用。ELISA测定小鼠脑匀浆液中γ-氨基丁酸(GABA)、谷氨酸(Glu)、谷氨酰胺(Gln)、5-羟色胺(5-HT)和5-羟吲哚乙酸(5-HIAA)含量。结果显示,与空白对照组比较,阈下剂量的戊巴比妥钠可使复方灵芝组小鼠入睡率极显著增加(P<0.01),阈上剂量的戊巴比妥钠可使复方灵芝组小鼠睡眠潜伏期极明显缩短(P<0.01)。与空白对照组比较,复方灵芝组小鼠脑内GABA含量极显著上升(P<0.01),Glu、Gln含量极显著下降(P<0.01)。结果表明复方灵芝可能通过增加小鼠脑内GABA含量,降低Glu、Gln含量,起到镇静、催眠的作用。To study the sedative and hypnotic effects of compound of Ganoderma lucidum on mice and the neurotransmitter contents in the brain,mice were randomly divided into blank control group[water,0.2mL/(10 g·bw)],positive drug group[estazolam,0.26mg/(10 g·bw)],and compound of Ganoderma lucidum group[126 mg/(10 g·bw),calculated by crude drug],with 10 mice in each group.Each group was given the relevant medicine intragastrically once a day,for consecutive 8 days.The mice were then given subthreshold doses and threshold doses of pentobarbital sodium to induce sleep,and their sleep status,sleep latency,and sleep duration were recorded.The contents ofγ-aminobutyric acid(GABA),glutamate(Glu),glutamine(Gln),5-hydroxytryptamine(5-HT)and 5-hydroxyindoleacetic acid(5-HIAA)in brain homogenate were determined by enzyme-linked immunosorbent assay.The results showed that compared with the blank control group,compound of Ganoderma lucidum group had an increased sleep rate and the duration of sleep,shortened incubation period of sleep after given subthreshold dose pentobarbital sodium(P<0.01),and an shorten sleep latency after given suprathreshold dose pentobarbital sodium.Compared with the blank control group,brain GABA levels in comprund of Ganoderma lucidum group significantly increased(P<0.01),while the contents of Glu and Gln remarkably decreased(P<0.01).The results indicate that compound of Ganoderma lucidum may play a calming and hypnotic role by increasing the content of GABA in the brain of mice and reducing the content of Glu and Gln.
