复方盐酸利多卡因凝胶剂中盐酸利多卡因和恩诺沙星含量的测定方法

摘　　要：为了建立用高效液相色谱(HPLC)法测定复方盐酸利多卡因凝胶剂中盐酸利多卡因和恩诺沙星含量的方法。采用盐酸利多卡因的色谱条件为C18柱(5μm,250 mm×4.6 mm),磷酸盐缓冲液-乙腈(50∶50)(用磷酸调节至pH 8.0)为流动相,进样量20μL,检测波长为254 nm;恩诺沙星的色谱条件为C18柱(5μm,250 mm×4.6 mm),0.025 mol/L磷酸溶液(用三乙胺调节至pH 3.0)-乙腈(83∶17)为流动相,进样量20μL,检测波长为278 nm。结果显示:盐酸利多卡因在0.1005~1.0049 mg/mL范围内线性关系良好,恩诺沙星在4.98~49.79μg/mL范围内线性关系良好,相关系数R分别为0.9999和0.9996,平均回收率分别为99.2%(RSD为0.77%)、100.3%(RSD为0.88%)。结果表明本试验所建立的HPLC法快速简便、准确可靠,可用于复方盐酸利多卡因凝胶剂中盐酸利多卡因和恩诺沙星的含量测定。This study aimed to establish a HPLC method to determine lidocaine hydrochloride and enrofloxacin contents of compound lidocacaine gel. For the chromatographic conditions to determine lidocaine hydrochloride, C18 chromatographic column was used. The mobile phase was phosphate buffer and acetonitrile(50∶50, adjusted to pH 8.0 with phosphoric acid). The injection volume was 20 μL, the detection wavelength was 254 nm. For the chromatographic conditions of determine enrofloxacin, C18 chromatographic column was used. The mobile phase was 0.025 mol/L phosphoric acid solution(adjusted to pH 3.0 with triethylamine) and acetonitrile(83∶17), injection volume was 20 μL, and the detection wavelength was 278 nm. The results showed that lidocaine hydrochloride and enrofloxacin all had a good linear relationship in the range of 0.100 5 ~1.004 9 mg/mL(R=0.999 9)and 4.98~49.79 μg/mL(R=0.999 6), respectively. The average recovery was 99.2% with RSD 0.77% and 100.3% with RSD 0.88%, respectively. The established HPLC methods are quick, simple, accurate and reliable, and could be applied to determine lidocaine hydrochloride and enrofloxacin contents of compound lidocacaine gel.
