摘 要:以红花烟草未分化幼苗为试验材料,研究了黑暗条件和光照条件(10h/d)下红花烟草叶片愈伤组织形成的差异。试验方法使用诱导培养基(MS+BA 0.20mg/L+NAA 0.20mg/L),分别从烟叶上获得愈伤组织。22天后将愈伤组织转移至分化培养基(MS+BA 0.80mg/L+NAA0.20mg/L)中,在24℃光照周期10h/d下观察芽分化2个月2天。结果表明,光照条件下愈伤组织诱导率较高,萌发率与光照条件有关,萌发系数较高,分化率与光照条件有关。本实验以红花烟草为材料,建立了植物组织培养再生体系。The influence of light on tobacco callus induction and plant regeneration was investigated using Nicotiana xsanderae indeterminate seedlings as experimental material and control light and darkness.The experimental method was to induce inoculation induction medium(MS+BA 0.20mg/L+NAA 0.20mg/L)under dark condition of 10h/d and 24 degree centigrade light condition,respectively callus were generated from tobacco leaves.Callus were transferred to differentiation medium(MS+BA 0.80mg/L+NAA 0.20mg/L)after 22 days,bud differentiation was observed for two months and two days under a 24 degree centigrade light cycle of 10h/d.The results showed the callus induction rate was higher under light condition,the germination rate was related to light condition,the germination coefficient was higher under light condition,and the differentiation rate was related to light condition.Because of the limited data and the absence of repeated experiments,it is preliminarily concluded that the induction of callus under light condition is more favorable than that under dark condition.
