摘 要:目的探讨电针水沟穴对脑梗死模型大鼠脑血管平滑肌钙样蛋白(Calponin)和钙调结合蛋白(Caldesmon)表达的影响。方法78只雌性Wistar大鼠采用随机数字表法分为空白组6只,模型组、电针组和假手术组各24只。采用腔内线栓法阻滞大鼠左侧大脑中动脉,制备脑梗死模型。电针组造模后即电针水沟穴20min,空白组、假手术组、模型组大鼠仅同样抓取固定。采用免疫印迹法检测各组大鼠造模后0.5、1、3、6h脑血管平滑肌Calponin和Caldesmon表达水平。结果与假手术组比较,模型组大鼠1、3、6h脑血管平滑肌Calponin表达量下调[(0.68±0.17)比(0.89±0.12)、(0.40±0.17)比(0.89±0.08)、(0.32±0.01)比(0.97±0.06),P均<0.05];与模型组比较,电针组大鼠3、6h脑血管平滑肌Calponin表达水平上调[(0.73±0.09)比(0.40±0.17)、(0.69±0.03)比(0.32±0.01),P均<0.05];与假手术组比较,模型组大鼠1、3、6h Caldesmon表达水平下调[(0.70±0.06)比(1.04±0.16)、(0.61±0.24)比(0.82±0.08)、(0.51±0.19)比(0.83±0.04),P均<0.05];与模型组比较,电针组大鼠1、3、6h Caldesmonn表达水平均上调[(0.80±0.10)比(0.70±0.06)、(0.73±0.06)比(0.61±0.24)、(0.60±0.06)比(0.51±0.19),P均<0.05]。结论电针水沟穴可抑制脑梗死模型大鼠脑血管平滑肌收缩关键蛋白Calponin和Caldesmon表达下调。Objective To determine the effect of electroacupuncture at Shuigou acupoint on the expression of Calponin and Caldesmon in cerebral vascular smooth muscle in a rat model of cerebral infarction.Methods Seventy-eight female Wistar rats were randomly divided into blank(n=6),model(n=24),electroacupuncture group(n=24),and sham operation groups(n=24).The cerebral infarction model was developed by blocking the rat left middle cerebral artery using intracavitary suture.Model rats were subjected to the electroacupuncture at Shuigou acupoint in the electroacupuncture group,while the blank,model and sham operation groups of rats were only grabbed and held in the same way as electroacupuncture group.After that,rat cerebral smooth muscle was isolated at 0.5,1,3,and 6 h after modeling and used for Western blot analysis of Calponin and Caldesmon expression.Results The data showed that the expression of calponin in cerebral smooth muscle was downregulated in the model rats compared with the sham operation group of rats at 1,3,and 6 h(0.68±0.17 vs.0.89±0.12,0.40±0.17 vs.0.89±0.08,and 0.32±0.01 vs.0.97±0.06;all P<0.05).In contrast,the expression of calponin in cerebral smooth muscle was increased in the electroacupuncture rats compared with that of the model rats at 3 and 6 h(0.73±0.09 vs.0.40±0.17 and 0.69±0.03 vs.0.32±0.01,respectively;P<0.05),whereas the expression of caldesmonn in the cerebral smooth muscle was downregulated in the model group of rats compared with the sham operation rats at 1,3,and 6 h(0.70±0.06 vs.1.04±0.16,0.61±0.24 vs.0.82±0.08,and 0.51±0.19 vs.0.83±0.04,respectively;all P<0.05).However,the expression of caldesmonn in cerebral smooth muscle was upregulated in the electroacupuncture group at 1,3,and 6 h(0.80±0.10 vs.0.70±0.06,0.73±0.06 vs.0.61±0.24,and 0.60±0.06 vs.0.51±0.19,respectively;all P<0.05).Conclusion Electroacupuncture at Shuigou acupoint could inhibit the downregulation of Calponin and Caldesmon in the cerebral smooth muscle in a rat model of cerebral infarction.
