摘 要:目的探索白花蛇舌草提取物多糖和总黄酮对于结肠癌细胞HT-29细胞的抑制作用。方法以白花蛇舌草全草提取多糖及总黄酮,通过MTT实验探索不同浓度的多糖及总黄酮对HT-29细胞增殖活性的影响,并观察不同浓度多糖及总黄酮对细胞形态改变及细胞核染色质的变化的影响,通过划痕实验研究多糖及总黄酮对HT-29细胞迁移能力的影响。结果白花舌草多糖及总黄酮对结肠癌HT-29细胞均具有毒性作用,抑制细胞的增殖,其抑制作用表现为量效关系;用不同浓度处理细胞后,随着药物浓度的升高,细胞逐渐缩小、变圆,胞质皱缩,细胞间间隙变大,呈现分散生长,当高浓度作用于细胞时,细胞脱壁,悬浮于培养液中;用DAPI溶液对细胞核进行染色,发现细胞核逐渐缩小,呈现出核固缩,部分表现为核碎裂;用50μg/ml多糖及5μg/ml的总黄酮作用于细胞进行划痕实验,当作用48 h后,细胞迁移度明显小于对照组。结论白花蛇舌草提取物多糖及总黄酮对于结肠癌HT-29细胞均具有抑制作用,抑制细胞增殖,促进凋亡,并抑制细胞迁移。Objective To investigate the inhibitory effect of polysaccharides and total flavonoids from Oldenlandia diffusa(OD)extracts on colorectal cancer HT-29 cells.Methods Polysaccharides and total flavonoids were extracted from the whole plants of OD.MTT assay was used to examine the effect of extracted polysaccharides and total flavonoids at different concentrations on the proliferation of HT-29 cells.The effect of extracted polysaccharides and total flavonoids at different concentrations on cell morphology and nuclear chromatin was observed.Scratch assay was used to measure the effect of polysaccharides and total flavonoids on migration of HT-29 cells.Results The extracted polysaccharides and total flavonoids from OD exhibited toxic effects on colorectal cancer HT-29 cells,and inhibited the cell proliferation in a dose-effect manner.Along with increasing concentrations of these agents in the cell treatment,the HT-29 cells gradually shrank in size and rounded in shape,showing wrinkled cytoplasm,enlarged intercellular spaces,and dispersed growth.Under high concentrations of the treatment agents,the cells were shed from cellular walls and suspending in the culture medium.DAPI staining of the cell nuclei showed that the cell nuclei gradually shrank in size,showing pyknosis,and even karyorrhexis in some of the cells.Scratch assay with 50μg/ml polysaccharides and 5μg/ml total flavonoids after treatment for 48 hours showed apparently lower cell migration as compared with the control group.Conclusion Polysaccharides and total flavonoids from Oldenlandia diffusa exhibit inhibitory effects on colorectal cancer HT-29 cells through inhibiting cell proliferation,promoting apoptosis,and suppressing cell migration.
