1型糖尿病大鼠睾丸内转录组差异表达基因分析

摘  要:目的探讨糖尿病大鼠睾丸的转录组学改变。方法 2019年9月至2020年5月从华中科技大学同济医学院实验动物中心购入18只4周龄SD大鼠,其中8只作为对照组大鼠,另外10只大鼠建立1型糖尿病大鼠模型,最终获得对照组与糖尿病组大鼠各6只。分别从两组中筛选出3只大鼠后,对6只大鼠的睾丸进行RNA测序,比较正常大鼠与1型糖尿病大鼠睾丸组织的转录组差异,并通过生物信息学分析对靶基因可能相关通路进行了预测以及实时定量聚合酶链反应实验验证。采用双样本双尾t检验进行统计分析。结果糖尿病组大鼠睾丸重量明显高于对照组[(0.42±0.15) g比(2.27±0.20) g,P<0.01,t=-18.124],睾丸生精小管结构受到严重损伤,糖尿病组精子质量低于对照组。糖尿病组睾丸共有138个基因高于对照组,119个基因低于对照组,其中4个基因与睾丸发育和精子的发生直接相关,可能参与了类固醇激素的合成以及氧化还原稳态的维持。2个基因与糖原合成、生长发育密切相关。经实时定量反转录聚合酶链反应(RT-qPCR)验证,以上基因的表达趋势与RNA测序结果一致。结论糖尿病可能通过影响睾丸内生精相关基因以及糖原代谢基因的转录表达,从而损害睾丸的生精功能。Objective To investigate the transcriptome changes of testis in rats with diabetes mellitus.Methods This study was conducted from May 2020 to September 2019.184-week-old SD rats were purchased from the Experimental Animal Center of Tongji Medical College of Huazhong University of Science and Technology,of which 8 were used as control rats,and the other 10 rats were used to establish the model of type 1 diabetes.Finally,6 rats in control group and 6 rats in diabetes group were obtained.The testes of 3 rats were selected from the two groups,which tobesequenced by RNA.Then we compared the difference of the transcriptional group between normal rats and type 1 diabetic rats.The possible related pathways of target genes were predicted by GO analysis and GESA analysis and verified by quantitative real-time polymerase chain reaction(RT-qPCR).All statistical tests were performed with two-sample two-tailed t tests.Results Compared with the control group,the testisweight of diabetic rats decreased significantly[(0.42±0.15)g vs.(2.27±0.20)g,t=-18.124,P<0.01],and the structure of testicular seminiferous tubules was seriously damaged,accompanied by a decline in sperm quality.A total of 138 genes were up-regulated and 119 genes were down-regulated.Fourgenes are directly related to testicular development and spermatogenesis,and may be involved in the synthesis of steroids.Two genes are closely related to steroid hormones biosynthesis,growth and development.The results from RT-qPCR clearly indicated that the expression of these genes was significantly decreased due to Diabetes.Conclusion Diabetes may damage the spermatogenic function of testis by affecting the transcriptional expression of spermatogenesis-related genes and glycogen metabolism genes in testes.
