摘 要:目的探讨哺乳动物雷帕霉素靶蛋白(mammalian target of rapamycin,mTOR)在小鼠毛囊早期发育中的表达与定位及其意义。方法本研究选取胚胎期14.5 d、15.5 d、16.5 d、17.5 d、18.5 d(E14.5、E15.5、E16.5、E17.5、E18.5)小鼠皮肤,采用免疫荧光染色技术,检测磷酸化的雷帕霉素靶蛋白(phosphorylate-mammalian target of rapamycin,p-mTOR)在小鼠胚胎期皮肤和毛囊发育过程中的表达与定位。结果在E14.5时期p-mTOR主要表达于表皮中。在E15.5~E16.5时期p-mTOR主要在表皮和毛芽中高度表达。在E17.5时期p-mTOR主要在表皮和毛钉中高度表达。在E18.5时期p-mTOR主要在表皮及未成熟的毛囊中高度表达;在E16.5、E17.5、E18.5时期,随着毛囊逐步发育,p-mTOR在小鼠早期毛囊中的表达水平差异无统计学意义(P>0.05),而p-mTOR在表皮层中的荧光强度逐渐减弱,差异有统计学意义(P<0.01);在E18.5时期p-mTOR在小鼠早期毛囊中的荧光强度到达最高且显著高于毛囊外表皮层的荧光强度,差异具有统计学意义(P<0.05)。结论mTOR的表达与毛囊早期发育有着十分密切的关系,可能在毛囊早期发育中起到一定的作用。Objective To detemine the expression and location of mTOR in the early development of mice hair follicles,and to explore its significance.Methods In this study,14.5 d,15.5 d,16.5 d,17.5 d,18.5 d(E14.5,E15.5,E16.5,E17.5,E18.5)of embryonic mice were selected,the expression and location of p-mTOR in the development of mice embryonic skin and hair follicles were determined by immunofluorescence.Results p-mTOR was mainly expressed in the epidermis during E14.5.During E15.5~E16.5,p-mTOR was mainly expressed in epidermis and hair germ.During E17.5,p-mTOR was highly expressed in the epidermis and hair peg.During E18.5,p-mTOR was mainly expressed in the epidermis and immature hair follicles.During the E16.5,E17.5,and E18.5 periods,as the hair follicles gradually developed,the expression of p-mTOR did not change in early mice hair follicles(P>0.05),while the fluorescence intensity of p-mTOR in the epidermal layer gradually decreased,and the difference was statistically significant(P<0.01).The fluorescence intensity of p-mTOR in mice hair follicles reached the highest at E18.5 and was significantly higher than that of the epidermal layer,and the difference was statistically significant(P<0.05).Conclusion The expression of mTOR is closely related to the early development of hair follicles,and mTOR plays critical role in the early development of hair follicles.
