摘　　要：目的研究泽泻95%乙醇提取物化学成分及抑制CYP1B1酶的活性。方法采用酶孵育反应实验测定泽泻醇提取物对CYP1B1酶的抑制活性,并通过超高效液相色谱串联离子阱飞行时间质谱(UPLC-IT-TOF/MS)技术,使用ESI离子源,Shimadzu Composition Formula Predictor软件定性分析活性物质。结果泽泻醇提取物具有显著抑制CYP1B1酶的活性,抑制率为93.96%。通过多级质谱数据分析,结合氮规则和Scifinder数据库检索,从泽泻醇提取物中鉴定了25个原萜烷型三萜类成分,可能是抑制CYP1B1酶的主要成分之一。结论本研究快速、准确分析了泽泻醇提取物中原萜烷型三萜类成分,为其药效物质基础和质量控制提供了依据。Objective To study the active chemical constituents in Alismatis Rhizome 95%ethanol extract inhibiting CYP1B1 enzyme.Methods The inhibition activity of Alismatis Rhizome ethanol extract toward CYP1B1 enzyme was tested through enzyme incubation reaction experiment.Chemical compositions of Alismatis Rhizome ethanol extract were identified by UPLC-IT-TOF/MS with ESI ion and Shimadzu Composition Formula Predictor software.Results Alismatis Rhizome ethanol extract showed the most significant inhibition with the inhibitory value of 93.96%toward CYP1B1 enzyme.Through the analysis of the multistage tandem mass spectrometry,combined with the application of nitrogen rule and the Scifinder database search,25 protostane-type triterpenes were identified.Conclusion This study quickly and accurately analyzed the protostane-type triterpenes in the Alismatis Rhizome ethanol extract,which provided the basis for the pharmacodyamic material basis and quality control.
