摘 要:为了体外研究游离脂肪酸(NEFA)对奶牛多形核中性粒细胞(PMN)跨奶牛乳腺上皮迁移功能的影响,用Transwell嵌套构建了跨乳腺上皮迁移模型,并以此模型研究了NEFA对PMN跨乳腺上皮迁移功能的影响。结果显示,不同浓度的油酸(OA)可显著促进PMN跨乳腺上皮迁移数量(P<0.01),其作用特点呈剂量依赖效应;棕榈酸(PA)及硬脂酸(SA)对PMN跨乳腺上皮迁移数量并无显著影响(P>0.05)。结果表明,NEFA不同组分对奶牛PMN迁移功能具有不同的调控作用,通过改善饲料成份和配比提高OA在奶牛体内NEFA中的占比,可能有助于防治奶牛乳腺炎。In order to investigate the effects of NEFA on the migration of neutrophils across the mammary in vitro,MAC-T were cultured in transwell basement membrane which was used as a trans-epithelial migration model and the effect of NEFA on the transepithelial migration of PMN was studied.The results showed that different concentrations of oleic acid(OA)could significantly promoted the PMN trans-epithelial migration in a dose-dependent manner(P<0.01),palmitic acid(PA)and stearic acid(SA)had no effect on the PMN trans-epithelial migration(P>0.05),suggesting that different components of NEFA have different regulatory effects on PMN migration function in dairy cows.It may be helpful to prevent and control mastitis by improving feed composition and increasing the proportion of OA in NEFA.
