摘  要:旨在研究母鼠长期铜暴露对子代大鼠睾丸功能的影响。将24只健康雌性SD大鼠随机分为A、B、C 3组,每组8只,A、B、C组分别饲喂AIN-93G基础日粮、AIN-93G基础日粮+CuSO4120 mg/kg、AIN-93G基础日粮+CuSO4240 mg/kg 3种不同铜水平日粮,试验期8周。雌鼠与性成熟雄性SD大鼠交配受孕后,在妊娠期和哺乳期继续铜覆盖。断乳后饲喂AIN-93G基础日粮,待子代雄性大鼠10周龄后,每组随机取8只,分别标记为A0、B0、C0组,记录体质量及睾丸、附睾脏器指数、检测睾丸组织铜水平、血清铜水平及抗氧化指标,计算精子数量及活力,HE染色观察睾丸生精小管结构。结果显示,随着铜水平增加,子代大鼠体质量下降;血清和睾丸中的铜增加;超氧化物歧化酶(SOD)、谷胱甘肽过氧化酶(GSH-Px)活性在亲代覆盖240 mg/kg铜组显著降低(P<0.05);大鼠血清丙二醛(MDA)水平随铜水平的增加而增加(P<0.01);日粮添加240 mg/kg铜的亲代其子代大鼠血清铜蓝蛋白(CP)显著高于日粮不添加铜的子代大鼠(P<0.05);睾丸生精细胞层数逐渐减少,各级生精细胞排列紊乱、数量减少或缺失,管腔内成熟的精子数量在亲代添加240 mg/kg铜的子代大鼠中显著减少(P<0.05)。结果表明,高铜诱导子代大鼠发生氧化应激,阻碍了大鼠的精子成熟和精子发生,导致生殖毒性。This article aims to study the effects of long-term copper exposure in female rats on the testis function of offspring rats.Twenty-four healthy female SD rats were randomly divided into A,B,and C three groups,with 8 rats in each group,each group was fed three diets with different copper levels:group A was fed with AIN-93 G basal diet,group B was fed with AIN-93 G basal diet+120 mg/kg CuSO4,and group C was fed with AIN-93 G basal diet+240 mg/kg CuSO4,respectively,the trial period was eight weeks.After mating with normal male SD rats for conception,the female rats continued to be fed with diets with copper during pregnancy and lactation.After weaning,they were fed with AIN-93 G basal diet,when the offspring male rats were 10 weeks old,eight rats were randomly selected from each group and labeled as A0,B0 and C0 groups and their body weight and the index of testis and epididymis were recorded,the copper levels of testicular tissue,serum copper levels and antioxidant indicators were detected,sperm count and vitality were determined,and the structure of testicular seminiferous tubules was observed by HE staining.The results showed that as the copper level increased,the weight of the offspring rats decreased and the content of copper in serum and testis increased;the activities of SOD and GSH-Px significant decreased in the parental generation group fed with 240 mg/kg copper(P<0.05);the serum MDA level of rats in each group increased with the increase of copper level(P<0.01);the serum CP was significantly higher than that of the offspring rats without adding copper in the diet(P<0.05);the number of spermatogenic cells in the testis gradually decreased,the arrangement of spermatogenic cells at all levels disordered,and the number was reduced;there was a significant decrease in the number of mature sperm in the lumen of the parental rats with 240 mg/kg copper(P<0.05).The above results indicate that high copper induces oxidative stress in offspring rats and hinders sperm maturation and spermatogenesis in mice,and it has r
