摘 要:[目的]观察补阳还五汤对间歇低氧大鼠胰腺内质网应激蛋白GRP78、Caspase-12、CHOP表达及超微结构的影响。[方法]24只SD大鼠随机分为常氧对照组、模型组、补阳还五汤组,每组8只,干预10周以后检测血清空腹血糖(FBG)、糖化血清蛋白(GSP)、空腹胰岛素(FINS)及胰腺内质网蛋白GRP78、Caspase-12、CHOP的表达情况,电镜观察胰腺组织结构。[结果]①与常氧对照组比较,模型组FBG、FINS水平明显上升(P<0.05);与模型组比较,补阳还五汤组FBG、FINS水平明显下降(P<0.05);而三组GSP水平无显著性差异。②与常氧对照组比较,模型组GRP78、CHOP、Caspase-12蛋白表达水平显著升高(P<0.05);与模型组比较,补阳还五汤组GRP78、CHOP、Caspase-12表达水平显著降低(P<0.05);且与常氧对照组GRP78、CHOP、Caspase-12水平相当,无明显差异(P>0.05)。③模型组胰腺细胞核膜不规则缺损,线粒体膜缺损,粗面内质网扩张;补阳还五汤组干预后线粒体排列较整齐,与模型组相比空泡变性、肿胀、嵴扩张减轻;粗面内质网轻度扩张、变形,排列较规律。[结论]补阳还五汤可通过减轻慢性间歇低氧大鼠胰腺内质网应激水平,良性调节GRP78、Caspase-12和CHOP蛋白含量,从而保护胰腺。[Objective]To observe the effect of Buyang Huanwu Decoction on the expression and ultrastructure of the stress proteins GRP78,caspase-12 and CHOP in the pancreas of intermittent hypoxia rats.[Methods]Twenty-four SD rats were randomly divided into oxygen-free control group,model group and Buyang Huanwu Decoction group,with 8 in each group.After 10 weeks of intervention,the expressions of FBG,GSP,FINS were tested,and GRP78,Caspase-12 and CHOP were detected.The structures of pancreatic tissue were observed under electron microscopy.[Results]①FBG and FINS in model group were significantly higher than that in normal oxygen control group(P<0.05).The FBG and FINS of Buyang Huanwu Decoction group were significantly lower than those of the model group(P<0.05).The GSP in the three groups showed no significance difference.②The expression level of GRP78,CHOP and Caspase-12 in the model group were significantly increased than those in the normoxic control group(P<0.05).Compared with the model group,the expression levels of GRP78,CHOP,and Caspase-12 in the Buyang Huanwu Decoction group were significantly reduced(P<0.05);and in the normoxic control group were similar,with no significant difference(P>0.05).③In the model group,the pancreatic cells had irregular nuclear membrane defect,mitochondrial membrane defect and rough endoplasmic reticulum expansion.After the intervention of Buyang Huanwu Decoction the arrangement of mitochondria was more orderly,compared with the model group,the vacuolar degeneration,swelling,and cristae expansion were reduced;the rough endoplasmic reticulum was slightly expanded and deformed,and the arrangement was more regular.[Conclusion]Buyang Huanwu Decoction can protect the pancreas by reducing the stress level of the pancreatic endoplasmic reticulum in rats with chronic intermittent hypoxia and positively regulating the Contents of GRP78,Caspase-12 and CHOP proteins.
