摘  要:目的:通过研究甘麦大枣汤对注意缺陷多动障碍(ADHD)模型动物自发性高血压大鼠(SHR)行为学变化、前额叶皮质层损伤的影响,探究本方治疗ADHD的作用机制与“脾藏意”理论的联系。方法:SHR大鼠随机分为甘麦大枣汤组(GMDZ组),利他林组(MPH组)和模型组(Model组),每组8只,另设8只韦斯达-京都大鼠(WKY)大鼠为空白对照组(Control组),适应性饲养7 d后,各组大鼠药物灌胃21 d。进行为期6 d的Morris水迷宫实验评估药物对SHR大鼠学习能力,冲动、多动行为的影响;通过苏木精-伊红(HE)染色、TUNEL染色观察大鼠前额叶皮质层病理形态变化。结果:空白对照组大鼠可见锥体细胞排列紧密,层次分明,结构有序,细胞形状规则,细胞核清晰可见,呈现深染,神经元细胞数量多,未观察到凋亡细胞;模型组大鼠见锥体细胞排列稀疏,层次不清,结构混乱,细胞形状不规则,细胞结构大多不清晰,神经元细胞数量较少,排列不齐,结构不完整,可见大量凋亡细胞;甘麦大枣汤组和利他林组可见锥体细胞排列较为紧密,层次比较分明,结构有序,细胞形状大多规则,细胞核多数清晰可见,呈现深染,神经元细胞数量尚可,排列比较整齐,结构较为完整,可见少量凋亡细胞。结论:甘麦大枣汤可以有效缓解ADHD模型SHR大鼠前额叶皮质层损伤,改善SHR大鼠关于ADHD的核心症状。Objective:To study the effect of Ganmai Dazao Decoction on behavioral changes and prefrontal cortex injury in spontaneously hypertensive rats(SHR)with attention deficit hyperactivity disorder(ADHD),and to explore the relationship between the mechanism of Ganmai Dazao Decoction in the treatment of ADHD and the theory of"spleen governing idea".Methods:SHR rats were randomly divided into Ganmai Dazao Decoction group(GMDZ group),methylphenidate group(MPH group)and model group,with 8 rats in each group.Another 8 Wistar Kyoto rats(WKY)were set as the blank control group.After adaptive feeding for 7 days,rats in each group were perfused with medicine for 21 days.Morris water maze test was conducted for 6 days to evaluate the effects of drugs on learning ability,impulse and hyperactivity in SHR rats;The pathological changes of prefrontal cortex were observed by hematoxylin eosin(HE)staining and TUNEL staining.Results:In the control group,pyramidal cells were closely arranged,well-defined,orderly in structure,regular in shape,clearly visible and deeply stained nuclei,and a large number of neuronal cells and no apoptotic cells were observed;In the model group,the pyramidal cells were sparsely arranged,the layers were unclear,the structure was chaotic,the cell shape was irregular,the cell structure was mostly unclear,the number of neuronal cells was small,the arrangement was uneven,the structure was incomplete,and a large number of apoptotic cells were seen;In GMDZ group and MPH group,pyramidal cells were closely arranged,with clear layers and orderly structure,most of the cells were regular in shape,most of the nuclei were clearly visible,showing deep staining,the number of neuronal cells was acceptable,the arrangement was relatively neat,the structure was relatively complete,and a small number of apoptotic cells could be seen.Conclusion:Ganmai Dazao decoction can effectively alleviate the injury of prefrontal cortex and improve the core symptoms of ADHD in SHR rats.
