摘 要:以叶艺兰(Cymbidium tracyanum var.‘huanghua’×C.iridioides D.Don.‘Yeyi’)叶片为试材,通过PCR技术,克隆CcMYB8基因全长,对其序列进行生物信息学、氨基酸同源性和亲缘关系分析,研究CcMYB8基因的功能,以期为揭示R2R3-MYB亚家族转录因子调控叶艺兰斑形成的机理研究奠定基础。结果表明:叶艺兰CcMYB8基因编码不稳定亲水性蛋白,具2个SANT保守结构域,全长为657 bp、编码219个氨基酸、分子量为24.974 kDa;与墨兰高度同源,其相似性达99.54%;系统进化树结果也表明,其与墨兰的亲缘关系最近;因此表明叶艺兰CcMYB8基因功能与该物种的相似或相同。The full length of CcMYB8 gene was cloned from the leaves of Cymbidium with variegated leaf(Cymbidium tracyanum var.‘huanghua’×C.iridioides D.Don.‘Yeyi’)by PCR,and its sequence was analyzed by bioinformatics,amino acid homology and genetic relationship,the function of the CcMYB8 gene were studied,in order to explain the regulation mechanism of R2 R3-MYB subfamily transcription factors on leaf spot formation.The results showed that the CcMYB8 gene of Cymbidium with variegated leaf was an unstable hydrophilic protein with a full length of 657 bp,encodes 219 amino acids,a molecular weight of 24.974 kDa and two SANT conserved domains,and was highly homologous to Cymbidium sinense,with a similarity of 99.54%.The phylogenetic tree also showed that it was closely related to Cymbidium sinense,so the function of CcMYB8 gene is similar or the same as that of this species.
