摘  要:目的探讨白芍总苷(TGP)对慢性肾小球肾炎(CGN)大鼠的肾脏保护作用及机制。方法通过注射阳离子化牛血清白蛋白(C-BSA)诱导CGN大鼠模型。将SD大鼠随机分成空白组、模型组、醋酸泼尼松组(5 mg·kg^(-1))及白芍总苷高、低剂量组(200、100 mg·kg^(-1)),灌胃给药,连续给药4周。给药结束后,检测大鼠24 h尿蛋白及血清肌酐(Scr)、尿素氮(BUN)水平;采用免疫荧光法检测肾脏组织中IgG和C3的沉积情况;Western Blot法检测肾脏组织中白细胞介素(IL)1β、转化生长因子(TGF)β、p62、TRAF6及核因子κB(NF-κB)蛋白表达情况。结果与空白组比较,模型组大鼠的24 h尿蛋白排泄量及血清中Scr、BUN含量显著升高(P<0.01),肾脏中IgG和C3沉积增多,IL-1β、TGF-β、p62、NF-κB及TRAF6蛋白表达明显上调(P<0.01)。与模型组比较,白芍总苷高、低剂量组大鼠的24 h尿蛋白排泄量及血清中Scr、BUN含量明显降低(P<0.05,P<0.01),肾脏组织中IgG和C3沉积明显减少,IL-1β、TGF-β、p62、NF-κB及TRAF6蛋白表达明显下调(P<0.05,P<0.01)。结论白芍总苷对CGN大鼠具有肾脏保护作用,其机制可能与下调IL-1β、TGF-β表达,影响p62/TRAF6/NF-κB信号通路激活有关。Objective To investigate the effect and the possible mechanism of total glycosides of paeony(TGP)on chronic glomerulonephritis model rats.Methods The model of chronic glomerulonephritis was induced by tail vein injection of cationized bovine serum albumin(C-BSA).SD rats were randomized into normal group,model group,high-,low-dose TGP groups(200,100 mg·kg^(-1)),and prednisone acetate group(5 mg·kg^(-1)).After the administration,the levels of urinary protein,serum creatinine(Scr)and urea nitrogen(BUN)in rats were detected.The deposition of IgG and C3 in the kidneys were detected by immunofluorescence.The expression levels of IL-1,TGF-β,p62,TRAF6,NF-κB protein in kidneys was detected by Western Blot.Results Compared with normal group,the levels of 24 h urinary protein,serum Scr and BUN in the model group were significantly increased(P<0.01),the deposition of IgG and C3 and the expression levels of IL-1β,TGF-β,p62,TRAF6,NF-κB protein in the model group increased(P<0.01).Compared with model group,the levels of 24-hour urinary protein,serum Scr and BUN in the high TGP group and low TGP group were significantly decreased(P<0.05,P<0.01);the deposition of IgG and C3 and the expression levels of IL-1β,TGF-β,p62,TRAF6,NF-κB protein in high TGP group and low TGP group were significantly decreased(P<0.05,P<0.01).Conclusion TGP exerts a therapeutic effect on chronic glomerulonephritis model rats by decreasing the expression of IL-1β,TGF-βand influencing activation of p62/TRAF6/NF-κB signaling pathway.
