菜粉蝶线粒体基因组序列测定与分析

摘　　要：测定了菜粉蝶(Pieris rapae)的线粒体基因组序列,分析了菜粉蝶线粒体基因组的结构特征。此外,结合GenBank已经公布的鳞翅目昆虫的线粒体基因组序列并采取最大似然法和贝叶斯法构建系统发育树,分析了粉蝶科属间的系统发育关系。菜粉蝶(P.rapae)线粒体基因组长度为15 106 bp (GenBank登录号为MW448362),包括22个tRNA基因、13个蛋白质编码基因、2个rRNA基因和一段控制区。菜粉蝶线粒体基因组具有较高的A+T含量(79.0%),其中13个蛋白质编码基因中UUA的相对密码子使用频率最高(2.91)。除trnS1之外,21个tRNA基因的二级结构皆是典型的三叶草结构。粉蝶科属间系统发育结果显示:确定小粉蝶属(Leptidea)为单系群,位于系统发育树的基部,分化较早;襟粉蝶属(Anthocharis)和鹤顶粉蝶属(Hebomoia)互为姐妹群;粉蝶属(Pieris)和飞龙粉蝶属(Talbotia)互为姐妹群,所构成的类群与云粉蝶属(Pontia)形成姐妹群;斑粉蝶属(Delias)与由绢粉蝶属(Aporia)和妹粉蝶属(Mesapia)构成的一支类群形成姐妹群,尖粉蝶属Appias单独成一支;迁粉蝶属(Catopsilia)和豆粉蝶属(Colias)聚为一支,与钩粉蝶属(Gonepteryx)形成姐妹群关系,黄粉蝶属(Eurema)单独成一支。In this study,we determined the mitochondrial genome sequence of Pieris rapae and analyzed the characteristics of genome structure.Furthermore,combined with the published mitochondrial genome sequences of Lepidoptera,we constructed the phylogenetic trees of Pieridae using maximum likelihood and Bayesian inference methods.The phylogenetic relationship among the genera of the family Pieridae was analyzed.The complete mitochondrial genome of Pieris rapae is 15106 bp in length(GenBank accession No.MW448362),including 13 protein-coding genes,22 tRNA genes,2 rRNA genes and a control region.The mitochondrial genome of Pieris rapae has a high A+T content(79.0%).The relative synonymous codon usage(RSCU)of UUA is the highest(2.91).All tRNA genes have the typical clover-leaf structure,but for that of trnS1.The results of phylogenetic analysis show that the Leptidea is a monophyletic group,located at the base of the phylogenetic tree,and differentiates earlier;the Anthocharis and the Hebomoia are sister groups;the Pieris and the Talbotia are sister groups,the constituent clusters form sister groups with the Pontia;the Delias form the sister groups with a cluster composed of the Aporia and Mesapia,the Appias forms a separate group;The Catopsilia and the Colias form a group and form a sister group relationship with the Gonepteryx,and the Eurema forms a separate group.
