摘 要:目的检测FXYD3在宫颈鳞癌患者组织中的表达,并分析其与患者临床病理参数的关系。方法分别通过GEPIA数据库及癌症基因组图谱(TCGA)门户网站UALCAN分析FXYD3在正常宫颈组织及宫颈鳞癌组织样本中的表达情况,使用cBioPortal在线软件预测分析FXYD3共表达基因,并通过ImageGP在线软件进行信号通路富集分析;STRING网站进行FXYD3蛋白互作共表达分析。收集2015年9月至2018年1月郑州市妇幼保健院就诊的65例宫颈鳞癌组织标本及50例因子宫肌瘤手术切除的正常宫颈组织标本,实时荧光定量PCR(qRT-PCR)检测各组FXYD3 mRNA的表达水平;免疫组化染色法检测FXYD3蛋白的表达水平,并分析其与宫颈鳞癌患者临床病理参数及预后的关系。结果数据库、qRT-PCR及免疫组化分析结果显示,宫颈鳞癌组织中FXYD3的表达水平[8.921(8.287,9.400)、10.244(9.569,10.749)、1.68±0.32、63.08%]均显著高于正常宫颈组织[0.995(0.346,5.837)、3.269(2.720,7.820)、1.00、28.00%],差异有统计学意义(Z分别为0.205、-6.107,t=15.011,χ^(2)=13.935;P均<0.05);FXYD3的表达与宫颈鳞癌组织类型、临床分期及淋巴结转移有关(χ^(2)分别为7.934,8.737,11.274,P均<0.05),FXYD3蛋白阳性表达患者的平均生存时间(OS)显著低于阴性表达患者(38.12个月vs 53.56个月,χ^(2)=5.485,P=0.019)。通过STRING网站在线绘制FXYD3蛋白互作网络图,并根据相关性评分分析证实相关性较强的前5个蛋白质分别为ATP1B1、ATP1B2、ATP1B3、ATP1A1、ATP1A2。通过cBioPortal数据库预测与FXYD3共表达基因417个;Enrichr网站通路富集分析结果显示,FXYD3共表达基因富集到的主要通路包括T细胞受体、B细胞受体、白细胞介素-17、趋化因子信号通路,氧化应激信号通路,非小细胞肺癌、膀胱癌等癌症信号通路,蛋白质水解、核黄素代谢、核酸合成代谢等信号通路。结论宫颈鳞癌组织中FXYD3的表达升高,且与患者组织类型、临床分期、淋�Objective To investigate the expression of FXYD3 in cervical squamous cell carcinoma(CSCC)and its correlation with clinicopathological parameters.Methods The expression levels of FXYD3 in normal cervical tissues and CSCC tissues were analyzed by the GEPIA database and the Cancer Genome Atlas(TCGA)portal UALCAN,respectively.The coexpressed genes of FXYD3 were predicted by the cBioPortal online software,and the signal pathway enrichment analysis was performed by the ImageGP online software.The interaction proteins of FXYD3 were analyzed on the STRING website.Sixty-five CSCC tissue samples and 50 normal cervical tissue samples from hysteromyoma patients were collected from our hospital during September 2015 and January 2018.The expression levels of FXYD3 mRNA were detected by real-time fluorescence quantitative PCR(qRT-PCR),and the expression levels of FXYD3 protein were determined by immunohistochemistry.Then,the correlations of expression levels of FXYD3 with clinicopathological parameters and prognosis of CSCC patients were analyzed.Results The results of database,qRT-PCR and immunohistochemistry analysis showed that the expression levels of FXYD3 in CSCC tissues(8.921[8.287,9.400],10.244[9.569,10.749],1.68±0.32,63.08%)were significantly higher than those in normal cervical tissues(0.995[0.346,5.837],3.269[2.720,7.820],1.00,28.00%;Z=0.205 and-6.107,t=15.011,χ^(2)=13.935;all P<0.05).The expression levels of FXYD3 were related to the tissue type,clinical stage and lymph node metastasis of CSCC(χ^(2)=7.934,8.737 and 11.274;all P<0.05).The mean overall survival(OS)of the patients with positive expression of FXYD3 was significantly lower than that of patients with negative expression(38.12 months vs 53.56 months,χ^(2)=5.485,P=0.019).The interaction networks of FXYD3 protein were drawn online through the STRING website.The correlation score analysis showed that the top five proteins with strong correlation were ATP1B1,ATP1B2,ATP1B3,ATP1A1 and ATP1A2,respectively.A total of 417 genes were predicted to be co-expresse
