摘  要:为深入分析HaNAC20转录因子在梭梭抗逆机制中的作用,以梭梭[Haloxylon ammodendron(C.A.Mey.)Bunge]HaNAC20转录因子为对象,克隆其编码基因,对其基本理化性质、所属亚族及同源基因进行生物信息学分析,使用qRT-PCR方法对HaNAC20在模拟干旱、高盐、模拟地表高温、IAA、ABA处理下的表达量进行分析,构建表达载体,进行亚细胞定位及转录激活活性分析。结果表明:克隆得到1个编码区全长为822 bp的梭梭NAC转录因子家族基因,命名为HaNAC20(登录号:KU845314),编码273个氨基酸。系统发育树显示HaNAC20属于NAC家族中与非生物胁迫相关的SNAC亚族。核定位信号预测及亚细胞定位结果表明HaNAC20定位于细胞核内。表达量分析结果显示:在模拟干旱、高盐、模拟地表高温、IAA、ABA处理下,HaNAC20表达量均显著上调。转录激活分析结果显示该转录因子C端具有转录激活活性。表明HaNAC20基因在梭梭响应非生物逆境胁迫、提高梭梭抗逆性中发挥重要作用。In order to further analyze the role of HaNAC20 transcription factor in the stress resistance mechanism of Haloxylon ammodendron(C.A.Mey.)Bunge ex Fenzl,HaNAC20 transcription factor from H.ammodendron was taken as object,its coding gene was cloned and its basic physicochemical properties,the subfamily and homologous genes were analyzed by bioinformatics methods.The expression of HaNAC20 under simulated drought,high salt,simulated high surface temperature,IAA and ABA treatment was analyzed by qRT-PCR.The expression vectors were constructed for subcellular localization and transcriptional activation activity analysis.The results showed that a gene of NAC transcription factor family of H.ammodendron with 822 bp coding region was cloned,named HaNAC20(accession number:KU845314),encoding 273 amino acids.Phylogenetic tree showed that HaNAC20 belonged to SNAC subfamily associated with abiotic stress in NAC family.Nuclear localization signal prediction and subcellular localization tests indicated that HaNAC20 was localized in the nucleus.The expression analysis showed that HaNAC20 expression was significantly up-regulated under simulated drought,high salt,simulated high surface temperature,IAA and ABA treatments.The results of transcriptional activation analysis showed that the C-terminal of this transcription factor had transcriptional activation activity.This study suggests that HaNAC20 gene plays an important role in response to abiotic stress and improve of the stres resistance of H.ammodendron.
