摘  要:对2008年-2015年分离的56个PRRSV毒株进行全基因测序和序列比对,并对主要易变区Nsp2、GP5和非编码区进行分析。全基因分析结果显示,56个PRRSV均为美洲型PRRSV,其中54个毒株为高致病性PRRSV变异株,另外2个为类NADC30 PRRSV。54个高致病性PRRSV变异株之间的同源性为95.2%~99.9%,与高致病性PRRSV变异株代表毒株JXA1的同源性为97.2%~99.3%。2个类NADC30分离株之间的同源性为99.9%,与NADC30 PRRSV代表毒株NADC30的同源性为95.4%。主要易变基因分析结果显示,非结构蛋白Nsp2和结构蛋白GP5仍在不断变异中。54个PRRSV变异毒株中有48个PRRSV变异株在非结构蛋白Nsp2发生30aa的不连续缺失,此外有6个高致病性PRRSV变异株在Nsp2区域还发生了其他缺失(其中1株多缺失12个核苷酸,1株多缺失19个核苷酸,2株多缺失29个核苷酸,1株多缺失48个核苷酸,1株多缺失120个核苷酸)。2个类NADC30 PRRSV非结构蛋白Nsp2存在131个核苷酸的不连续缺失。大部分分离株在重要结构蛋白GP5发生aa突变,尤其是糖基化位点的位置和数量发生了不同程度的变化。The whole sequences of 56 PRRSV isolated in China during 2008 to 2015 were determined,and then compared with representative PRRSV strains.Genetic variations of important genes Nsp2,GP5 and Non-coding region were analyzed.The results showed that 56 isolate belongs to American PRRSV.With in the 56 American PRRSVs,there were two NADC30-like PRRSVs and 54 HP-PRRSVs.The 54 HP-PRRSV isolates show 95.2%-99.9%homology,and they share 97.2%-99.3%homology comparing with representative strain of HP-PRRSV(JXA1).The homology between two NADC30-like strains was 99.9%,and their genome shares 95.4%similarity comparing with NADC30 PRRSV representative strain(NADC30).Gene sequence analysis of important viral proteins showed that PRRSV is changing constantly.Among 54 PRRSV variant strains,48 PRRSV variant strains have a 30aa non-contiguous deletion in the nonstructural protein Nsp2.In addition,6 highly pathogenic PRRSV variants also have other deletions in the Nsp2 region(1 strain additionally lacks 12 aa,1 strain additionally lacks 19 aa,2 strains additionally lack 29 aa,1 strain additionally lacks 48 aa,1 strain additionally lacks 120 aa).Two NADC30-like PRRSVs have a 131aa non-contiguous deletion in the nonstructural protein Nsp2.In the important structural protein GP5,most of the isolates show significant changes of GP5 gene,especially of the location and number of glycosylation sites.
