摘 要:目的:研究藏药镰形棘豆幼苗在不同B波段紫外线(ultraviolet radiation B,UVB)辐射下类黄酮的含量变化及黄酮合成相关酶的活性变化,从细胞水平探讨镰形棘豆类黄酮积累受UVB胁迫的影响及调节机制。方法:以镰形棘豆人工培养幼苗为实验材料,设置低辐照强度Q1(约2.0 W/m^(2))组、高辐照强度Q2(约3.0 W/m^(2))组及对照组,分别在UVB处理0、1、2、3、5、7 d后取样,测定总黄酮、鼠李柠檬素含量及苯丙氨酸解氨酶(phenylalanine ammonialyase,PAL)、查尔酮合酶(chalcone synthase,CHS)及查尔酮异构酶(chalcone isomerase,CHI)的活性。结果:UVB辐射能够促进镰形棘豆总黄酮和鼠李柠檬素的含量升高。UVB能够提高PAL的活性,增强效果Q1优于Q2。Q1剂量的UVB能够提高CHS和CHI的活性,Q2剂量的UVB短时间辐射能够提高CHS和CHI的活性,长时间辐射降低CHS和CHI的活性。结论:UVB辐射对镰形棘豆类黄酮物质的积累有明显的促进作用。低剂量的UVB能够刺激镰形棘豆黄酮合成相关酶的活性升高,促进类黄酮的生物合成。高剂量的UVB长时间辐射可能对镰形棘豆幼苗造成不可修复的损伤,不利于类黄酮的积累。Objective:To study the contents of flavonoid and activity of flavonoid synthesis related enzymes under different UVB radiation in the seedlings of Tibetan medicine Oxytropis falcate Bunge,and to explore the effects of UVB stress on flavonoid accumulation and its regulation mechanism at cellular level.Methods:After using cultured seedlings of Oxytropis falcate Bunge as experimental materials,they were allocated to low irradiation intensity Q1(about 2.0 W·m^(2))group,high irradiation intensity Q2(about 3.0 W·m^(2))group and the control group.After 0,1,2,3,5 and 7 days of UVB treatment,the contents of total flavonoids,rhamnetin and the activities of phenylalanine ammonialyase(PAL),chalcone synthase(CHS)and chalcone isomerase(CHI)were determined.Results:UVB radiation could increase the contents of total flavonoids and rhamnocitrin in Oxytropis falcate Bunge.UVB radiation can improve the activity of PAL and Q1 is better than Q2.Q1 dose of UVB can improve the activity of CHS and CHI,UVB irradiation at a dose of Q2 could raise the activity of CHS and CHI for a short time,it could reduce the activity of CHS and CHI for a long time.Conclusion:UVB radiation can obviously promote the accumulation of flavonoids in Oxytropis falcate Bunge.Low dose of UVB radiation could stimulate the activity of flavonoid synthesis related enzymes in Oxytropis falcate Bunge and promote the biosynthesis of flavonoids.high dose of UVB radiation for a long time may cause irreparable damage to Oxytropis falcate Bunge seedlings,which is not conducive to the accumulation of flavonoids.
