摘　　要：目的探讨子痫前期(preeclampsia,PE)患者胎盘组织中骨保护素(osteoprotegerin,OPG)和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的表达及其与疾病发展的相关性。方法通过qRT-PCR、Western blot和免疫组化法检测60例PE患者(试验组)和36例正常妊娠女性(对照组)胎盘组织中OPG和TNF-α基因mRNA及蛋白表达水平,并分析其与疾病发展的相关性。结果 PE患者胎盘组织中OPG基因mRNA及蛋白表达水平显著低于对照组(P<0.05),而TNF-α基因mRNA及蛋白表达水平则显著高于对照组(P<0.05);OPG与TNF-α蛋白在PE患者胎盘组织中的表达呈负相关(rs=-0.702,P<0.05)。结论 PE患者胎盘组织中OPG表达下调,TNF-α表达上调,二者表达呈负相关性。本研究为阐明PE的发病机制及探讨诊断和预后指标提供了实验依据。Objective To investigate the expressions of osteoprotegerin(OPG) and tumor necrosis factor(TNF)-α in placental tissue of patients with preeclampsia(PE)and their correlation with disease progression.Methods The mRNA and protein expression levels of OPG and TNF-α in placenta of 60 patients with PE(trial group)and 36 normal pregnant women(control group)were determined by qRT-PCR,Western blot and immunohistochemical assay,and the correlation between OPG/TNF-α expression and disease progression was analyzed.Results The expressions of OPG mRNA and protein in placental tissue of patients with PE were significantly lower,while those of TNF-α were significantly higher,than those in control group(each P<0.05).The OPG expression in placental tissue of patients with PE was negatively correlated with that of TNF-α(rs =-0.702,P<0.05).Conclusion The OPG expression in placental tissue of patients with PE was down-regulated,while that of TNF-α expression was up-related,which was negatively regulated.The study provided an experimental basis for understanding the pathogenic mechanism and investigating the indexes for diagnosis and prognosis of PE.
