益智清心颗粒对自发性高血压大鼠主要安全性指标的影响

摘　　要：目的观察益智清心颗粒对自发性高血压大鼠(SHR)主要安全性指标溶血相关指标、肝肾功能指标的影响。方法将Wistar Kyoto(WKY)大鼠设置为对照组,将SHR大鼠随机分为模型组、厄贝沙坦+多奈哌齐组、益智清心颗粒低剂量组、益智清心颗粒中剂量组和益智清心颗粒高剂量组,每组12只,雌雄各半。给药11周后检测外周血红细胞、血红蛋白、胆红素、肝肾功能指标等。结果红细胞相关指标:与对照组比较,模型组红细胞分布宽度降低(P<0.01);与模型组比较,益智清心颗粒低剂量组平均血红蛋白浓度降低(P<0.05)。胆红素相关指标:与对照组比较,模型组间接胆红素水平升高(P<0.05);与模型组比较,益智清心颗粒高剂量组间接胆红素、直接胆红素、总胆红素水平降低(P<0.05或P<0.01)。肝功能相关指标:与对照组比较,模型组天门冬氨酸氨基转移酶、γ谷氨酰转肽酶水平升高(P<0.01)。肾功能相关指标:与对照组比较,模型组尿素氮、尿酸水平升高(P<0.01)。结论益智清心颗粒对SHR无明显肝肾损伤,同时未见明显溶血反应。Objective To observe the effects of Yizhi Qingxin Granules on the main safety indexes in spontaneously hypertensive rats(SHR).Methods Wistar Kyoto(WKY)rats were select as the control group,SHR were randomly divided into model group,irbesartan+donepezil group,Yizhi Qingxin Granules low-dose group,Yizhi Qingxin Granules medium-dose group,and Yizhi Qingxin Granules high-dose group,with 12 rats in each group,and half male and half female.After 11 weeks of administration,peripheral blood red blood cells,hemoglobin,bilirubin,liver and kidney function indexes were detected.Results Compared with the control group,the red blood cell distribution width in the model group decreased(P<0.01);compared with the model group,the average hemoglobin concentration in the Yizhi Qingxin Granules low-dose group decreased(P<0.05).Compared with the control group,the level of indirect bilirubin in the model group increased(P<0.05);compared with the model group,the levels of indirect bilirubin,direct bilirubin and total bilirubin in the Yizhi Qingxin Granules medium-dose group decreased(P<0.05 or P<0.01).Compared with the control group,the levels of aspartate aminotransferase andγ-glutamyltranspeptidase in the model group increased(P<0.01).Compared with the control group,the levels of urea nitrogen and uric acid in the model group increased(P<0.01).Conclusion Yizhi Qingxin Granules used for SHR without obvious liver and kidney damage,and there were no obvious hemolysis reaction.
