摘 要:目的观察β-淀粉样蛋白1-40(Aβ1-40)对脑微血管内皮细胞活力、咬合蛋白及紧密连接蛋白1表达的影响。方法将细胞分为对照组、Aβ诱导组及不同剂量黄芪提取物组。采用噻唑蓝法测定细胞活力;蛋白印迹法检测咬合蛋白和紧密连接蛋白1表达;逆转录-集合酶链式反应(RT-PCR)分析咬合蛋白和紧密连接蛋白1 mRNA的表达。结果与对照组比较,Aβ诱导组细胞活力随着孵育天数增加逐渐下降(P<0.05),咬合蛋白和紧密连接蛋白1表达降低(P<0.05),咬合蛋白mRNA和紧密连接蛋白1 mRNA表达降低(P<0.05);与Aβ诱导组比较,黄芪提取物组细胞活力随着孵育天数增加逐渐升高(P<0.05),咬合蛋白和紧密连接蛋白1蛋白表达增加(P<0.05),咬合蛋白mRNA和紧密连接蛋白1 mRNA表达升高(P<0.05)。结论黄芪提取物能改善Aβ1-40损伤的脑部微血管壁内皮细胞活力,升高咬合蛋白和紧密连接蛋白1蛋白及mRNA表达,达到保护血脑屏障的目的。Objective To observe the effects ofβ-amyloid 1-40(Aβ1-40)on viability of brain microvascular endothelial cell,occlusal protein and zonula occludens-1.Methods The cells were divided into control group,Aβ-induced group,and different dose Astragalus extract group.Cell viability was detected by thiazoline-blue method.The expression of occlusal protein and zonula occludens-1 was detected by Western Blotting.The mRNA expressions of occlusal protein and zonula occludens-1 were detected by reverse transcription-aggregate enzyme chain reaction(RT-PCR).Results Compared with the control group,the cell viability of Aβ-induced group decreased gradually with the increase of incubation days(P<0.05),and the expressions of occlusal protein and zonula occludens-1(P<0.05)decreased.The mRNA expressions of occlusal protein and zonula occludens-1 decreased(P<0.05).Compared with the Aβ-induced group,the cell viability of Astragalus extract group was gradually increased with the incubation days increasing(P<0.05).The protein expressions of occlusal protein and zonula occludens-1 increased(P<0.05),and the mRNA expressions of occlusal protein and zonula occludens-1 increased(P<0.05).Conclusion Astragalus extract could improve the activity of endothelial cells in cerebral microvascular wall after Aβ1-40 injury,and increase the expression of occlusal protein and zonula occludens-1 and the mRNA of occlusal protein and zonula occludens-1,so as to protect the blood-brain barrier.
