摘  要:目的:探讨红花提取物对高糖处理的小鼠足细胞增殖、凋亡和炎症因子分泌的影响及分子机制。方法:将小鼠足细胞MPC5分为对照(NC)组、高糖(HG)组、不同浓度红花提取物组、红花提取物+HG组、miR-NC+HG组、miR-516a-5p+HG组、anti-miR-NC+HG组、anti-miR-516a-5p+HG组、红花提取物+miR-NC+HG组、红花提取物+miR-516a-5p+HG组。MTT检测细胞活性;流式细胞术检测细胞凋亡;RT-qPCR检测miR-516a-5p表达水平;ELISA检测白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平。结果:红花提取物处理后,高糖处理的小鼠足细胞中细胞活性升高,细胞凋亡率降低,IL-1β、IL-6、TNF-α水平降低,miR-516a-5p表达水平降低(P<0.05)。过表达miR-516a-5p促进高糖处理的小鼠足细胞炎症因子表达和细胞凋亡,抑制细胞增殖;抑制miR-516a-5p表达与其作用相反。miR-516a-5p过表达可以逆转红花提取物对高糖处理的MPC5细胞增殖、凋亡,炎症因子表达的影响。结论:红花提取物可能通过下调miR-516a-5p表达抑制高糖处理的小鼠足细胞炎症因子表达和细胞凋亡,促进细胞增殖。Objective:To explore the effect and molecular mechanism of safflower extract on the proliferation,apoptosis and secretion of inflammatory factors of mice podocytes treated with high glucose.Methods:The mouse podocyte MPC5 was divided into control(NC)group,high glucose(HG)group,different concentrations safflower extract group,safflower extract+HG group,miR-NC+HG group,miR-516 a-5 p+HG group,anti-miR-NC+HG group,anti-miR-516 a-5 p+HG group,safflower extract+miR-NC+HG group,safflower extract+miR-516 a-5 p+HG group.Methyl thiazolyl tetrazolium assay(MTT)detection of cell activity;flow cytometry detection of apoptosis;real-time fluorescence quantitative PCR(RT-qPCR)detection of expressions of miR-516 a-5 p;ELISA detects the levels of interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α).Results:After safflower extract treatment,the activity of cells in podocytes of mice treated with high glucose was increased,the apoptosis rate was decreased,the levels of IL-1β,IL-6,TNF-αwere decreased,and the expression of miR-516 a-5 p was decreased(P<0.05).Overexpression of miR-516 a-5 p promotes the expression of inflammatory factors and apoptosis in podocytes of mice treated with high glucose,and inhibits cell proliferation;inhibiting the expression of miR-516 a-5 p has the opposite effect.Overexpression of miR-516 a-5 p can reverse the effects of safflower extract on the proliferation,apoptosis and expression of inflammatory factors of MPC5 cells treated with high glucose.Conclusion:Safflower extract may inhibit the expression of inflammatory factors and apoptosis in podocytes of mice treated with high glucose,and promote cell proliferation by down-regulating the expression of miR-516 a-5 p.
