摘 要:共存离子是否直接干扰纤维素酶活力或者通过干扰还原性糖DNS法测定间接干扰纤维素酶活力的测定尚存在争议。通过外加不同种类的共存离子,研究了不同共存离子对DNS法测定葡萄糖标准液和纤维素酶活力的影响,并对共存离子干扰D N S法测葡萄糖含量及纤维素酶活性的原因进行了实验探索。实验结果表明:Mn^(2+)、Cu^(2+)、Fe^(3+)等金属离子对DNS法测定葡萄糖含量、DNS测定纤维素酶活力均产生不同程度的干扰,且浓度越高,干扰越大。当金属离子浓度为10mmol/L时,Mn^(2+)、Cu^(2+)分别导致葡萄糖的测定值为标准样的203.7%、24.3%,其余几种金属离子对测定值干扰较小。当Mn^(2+)、Fe^(3+)、Cu^(2+)浓度为10 mmol/L时,分别导致纤维素酶活性的测定值为实际值的207.9%、85.6%和83.1%。金属离子对DNS法测定纤维素酶活力的干扰机理,可能是由于金属离子自身的氧化还原性导致吸光度的变化以及金属离子对酶水解效率的影响等多种作用协同的结果。It is debatable whether coexisting ions directly interfere with determining result of cellulase activity or indirectly interfere with the determination result of cellulase activity by hindering reducing sugar determination.By adding different kinds of coexisting ions,the effects of different coexisting ions on the determination of glucose standard solution by DNS method werestudied,and the reasons for the interference of coexisting ions on the determination of glucose content and cellulase activity by DNS method were explored.The results showed that metal ions,such as Mn^(2+),Cu^(2+),Fe^(3+),significantly interfered with the DNS determination results of glucose content and cellulase activities.And the higher the iron concentration,the greater the interference.When the concentration is at 10 mmol,Mn^(2+)and Cu^(2+)lead to the determination of glucose determination being 203.7%and 24.3%of the standard sample respectively.The other metal ions have little influence on the determination result.When the concentrations of Mn^(2+),Fe^(3+)and Cu^(2+)were 10 mmol,the measured values of cellulase activity were 207.9%,85.6%and 83.1%of the actual values respectively.The interference of metal ions to the determination of cellulase activity by DNS method may be due to the change of absorbance caused by the oxidation or reduction of metal ions and the effect of metal ions on the hydrolysis efficiency.
