摘  要:为了探索禽网状内皮组织增生病病毒(reticuloendotheliosis virus,REV)感染对SPF雏鸡外周血液B淋巴细胞功能和免疫球蛋白含量变化的影响,试验将100只1日龄SPF雏鸡随机分为REV感染组和对照组,REV感染组(50只)雏鸡通过腹腔注射REV稀释液(0.5 mL/只),对照组(50只)雏鸡相同途径给予等量灭菌生理盐水,于REV感染后第1,7,14,21,28,42天,每组随机抽取5只,心脏无菌采血,应用细胞培养技术结合MTT和免疫学等检测方法,检测REV感染对SPF雏鸡外周血液B淋巴细胞增殖能力和IgG、IgA、IgM含量的影响。结果表明:SPF雏鸡感染REV后1~42 d,外周血液中无论是B淋巴细胞对脂多糖(LPS)的增殖能力还是血清IgG、IgA、IgM含量均不同程度低于对照组雏鸡,其中REV感染组B淋巴细胞对LPS的增殖能力在REV感染后第7,14天显著或极显著低于对照组雏鸡(P<0.05或P<0.01);IgG含量在REV感染后第14,28天显著低于对照组雏鸡(P<0.05);IgA含量在REV感染后第21,28,42天显著或极显著低于对照组(P<0.05或P<0.01);IgM含量在REV感染后第42天极显著低于对照组(P<0.01)。说明REV感染可导致SPF雏鸡外周血液的B淋巴细胞功能下降和免疫球蛋白IgG、IgA、IgM含量下降。In order to explore the effect of avian Reticuloendotheliosis virus(REV)infection on peripheral blood B lymphocyte function and immunoglobulin content of SPF chicks,1001-day-old SPF chicks were randomly divided into REV infection group and control group.The chicks of the REV infection group(50 individuals)were injected with REV diluent 0.5 mL/bird through the intraperitoneal cavity;the chicks of the control group(50 individuals)were given the same amount of sterilized saline in the same way.On 1,7,14,21,28 and 42 days after REV infection,5 chicks were randomly selected from each group,and blood was collected from the heart aseptically.Cell culture technology combined with MTT and immunological detection methods were used to detect the effect of REV infection on the proliferation of B lymphocytes and IgG,IgA,IgM in the peripheral blood of SPF chicks.The results showed that from 1 to 42 days after SPF chicks were infected with REV,both the proliferation ability of B lymphocytes to lipopolysaccharide(LPS)and the serum IgG,IgA and IgM levels in the peripheral blood were lower than those of the control chicks.Among them,the proliferation ability of B lymphocytes in the REV infection group on LPS was significantly or extremely significantly lower than that of the control chicks on 7,14 days after REV infection(P<0.05 or P<0.01);the content of IgG was significantly lower than those of control chicks at 14,18 days after virus infection(P<0.05),the content of IgA was significantly or extremely significantly lower than those of control chicks at 21,28,42 days after virus infection(P<0.05 or P<0.01);the content of IgM was significantly lower than those of control chicks at 42 days after virus infection(P<0.05);The results suggested that REV infection could cause the decline of B lymphocyte function in peripheral blood of SPF chicks and the decrease of immunoglobulin IgG,IgA,and IgM levels.
