摘  要:目的:探讨传染性单核细胞增多症患儿外周血中干扰素调节因子3(IRF-3)及miRNA-146a的表达水平及miRNA-146a对IRF-3表达的影响。方法:体内实验通过选取传染性单核细胞增多症患儿45例和健康对照者34例,运用Real-timePCR法检测外周血IRF-3和miRNA-146a的表达水平。体外实验通过转染miRNA-146a模拟物及其抑制剂检测miRNA-146a对IRF-3表达水平的影响。结果:与健康对照组相比,传染性单核细胞增多症患儿外周血IRF-3基因表达显著降低,差异有统计学意义(t=30.340,P<0.001),而且传染性单核细胞增多症患儿外周血miRNA-146a表达较健康对照组明显升高(t=34.659,P<0.001)。传染性单核细胞增多症患儿外周血IRF-3和miRNA-146a两者存在负相关性(r=-0.960,P<0.05)。在HeLa细胞中,转染miRNA-146a模拟物明显降低IRF-3 mRNA(t=8.270,P<0.001)及蛋白(t=46.170,P<0.001)表达,而miRNA-146a抑制剂明显上调IRF-3 mRNA(t=8.582,P<0.001)及蛋白(t=25.891,P<0.001)表达。结论:miRNA-146a可能参与EB病毒下调传染性单核细胞增多症患儿IRF-3基因的表达,而且参与传染性单核细胞增多症的发病。Objective:To investigate the expression levels of interferon regulatory factor 3(IRF-3)and miRNA-146a in peripheral blood of children with infectious mononucleosis and to explore the effect of miRNA-146a on IRF-3 expression.Methods:In the in vivo experiment,45 children with infectious mononucleosis and 34 healthy controls were selected to detect the expression levels of IRF-3 and miRNA-146a in peripheral blood by real-time PCR.In the in vitro experiment,the effect of miRNA-146a on IRF-3 expression was detected by transfection of miRNA-146a mimics and their inhibitors.Results:The expression of IRF-3 gene in peripheral blood of the healthy control group and the group of children with infectious mononucleosis was significantly different(t=30.340,P<0.001),while the expression of miRNA-146a in the group of children with infectious mononucle-osis was significantly higher than that of the healthy control group(t=34.659,P<0.001).There was a negative correlation(r=-0.960,P<0.05)between IRF-3 and miRNA-146a in peripheral blood of children with infectious mononucleosis.Overexpression of miRNA-146a can significantly reduce IRF-3 mRNA(t=8.270,P<0.001)and protein(t=46.170,P<0.001)expression,while miRNA-146a inhibitors can up-regulate IRF-3 mRNA(t=8.582,P<0.001)and protein(t=25.891,P<0.001)expression in HeLa cells.Conclusion:The expression of miRNA-146a may be involved in the negative regulation of IRF-3 by EB vi-rus in the patients with infectious mononucleosis and involved in the development of infectious mononucleosis.
