摘  要:目的研究儿咳灵糖浆对支气管哮喘的作用机制与Caspase-3、TNF-α蛋白的表达关系。方法将50只清洁级小鼠随机分成正常对照组、模型组、儿咳灵糖浆低剂量组、儿咳灵糖浆高剂量组、地塞米松组,通过致敏激发建立小鼠哮喘模型。采用HE染色和天狼星红病理学检测观察肺脏形态变化,免疫印迹法检测肺组织中Caspase-3、TNF-α的表达。结果与正常对照组相比,模型组小鼠肺泡细胞中出现大量炎性细胞浸润以及肺部组织纤维化,儿咳灵糖浆可以改善病理损伤。正常对照组小鼠支气管厚度和平滑肌厚度均低于模型组(P<0.05)。低剂量组和高剂量组支气管厚度和平滑肌厚度均低于模型组(P<0.05),且高剂量组低于低剂量组(P<0.05)。地塞米松组和高剂量组的支气管厚度、平滑肌厚度比较无统计学差异(P>0.05)。正常对照组小鼠血清中IFN-γ、IL-12、IL-6、TNF-α和肺组织中Caspase-3、TNF-a蛋白表达水平低于模型组(P<0.05)。低剂量组和高剂量组上述指标表达水平均低于模型组(P<0.05),且高剂量组低于低剂量组(P<0.05)。地塞米松组和高剂量组上述指标比较无统计学差异(P>0.05)。结论儿咳灵糖浆给药能有效降低小鼠肺组织的损伤以及Caspase-3和TNF-α的蛋白表达,从而发挥支气管哮喘模型小鼠的气道保护作用。Objective To study the mechanism of Erkeling syrup on treating bronchial asthma and the expression rela⁃tionship between caspase-3 and TNFα.Methods Fifty BABL/c mice were randomly divided into normal group,mod⁃el group,low-dose Erkeling syrup group,high-dose Erkeling syrup group,and dexamethasone group(10 mice/group).Through sensitization(OVA,aluminum hydroxide),stimulation(OVA,NS)to establish a mouse asthma model.24 hours after the last challenge,the mice were dissected to obtain alveolar lavage fluid(BALF)and lung tissue.The mor⁃phological changes of the lungs were observed by HE staining and Sirius red pathological examination,and the expres⁃sion of Caspase-3 and TNF-αin lung tissues was detected by Western-blot.Results Compared with the normal con⁃trol group,there were a large number of inflammatory cell infiltration and pulmonary fibrosis in the alveolar cells of the model group.Erkeling syrup can improve these pathological injuries.The bronchial thickness and smooth muscle thickness in the normal control group were lower than those in the model group(P<0.05).The bronchial thickness and smooth muscle thickness in both low-dose group and high-dose group were lower than those in model group(P<0.05),and those in high-dose group were lower than those in low-dose group(P<0.05).There was no significant dif⁃ference in bronchial thickness and smooth muscle thickness between dexamethasone group and high-dose Erkeling syrup group(P>0.05).IFN-γ,IL-12,IL-6,TNF-αin serum and Caspase-3,TNF-αin lung tissue of normal control group were lower than those in model group(P<0.05).The expression levels of above indicators in both low-dose group and the high-dose group were lower than the model group(P<0.05),and the high-dose group was lower than the low-dose group(P<0.05).There was no significant difference in above indicators between the dexamethasone group and the high-dose Erkeling syrup group(P>0.05).Conclusion Erkeling syrup can effectively reduce the injury of lung tissue,and the expression of Caspase-3 and
