黄樟素型沉水樟茎段组织培养研究

摘　　要：为保护和开发利用黄樟素型沉水樟,以黄樟素型沉水樟枝条为材料,研究外植体的采集时间、消毒时间和生长调节剂对黄樟素型沉水樟茎段组织培养的影响。结果表明:5月份采集黄樟素型沉水樟一年生半木质化枝条,修剪成带芽茎段,用质量浓度0.1%的HgCl_(2)消毒5 min,茎段的萌芽率高、污染率和褐化率低,茎段萌芽最适培养基为MS+0.8 mg/L 6-BA+0.15 mg/L IBA,萌芽率为66.67%;组培苗最适增殖培养基为MS+1.0 mg/L 6-BA+0.10 mg/L IBA,增殖系数为3.94;生根适宜培养基为1/2 MS+1.5 mg/L NAA,生根率81.60%。组培苗生根培养11~15 d,移入大棚内炼苗7~10 d,移栽到营养钵,移栽成活率可达90%以上。The effects of explant collection time,disinfection time and growth regulator on tissue culture of branches of Cinnamomum micranthum ct.safrole were studied,aiming to protect and exploit of C.micranthum ct.safrole.The results showed that the axillary bud germination rate of stem segments from semi-lignified stem segments of C.micranthum ct.safrole was highest with 0.1%HgCl_(2) disinfected for 5 min in the May,and the browning rate and pollution rate were low.The optimum medium for stem segment germination of C.micranthum ct.safrole was MS+0.8 mg/L 6-BA+0.15 mg/L IBA,the germination rate was 66.67%.The optimum medium for multiplication was MS+1.0 mg/L 6-BA+0.10 mg/L IBA,the multiplication coefficient was 3.94.The optimum medium for rooting was 1/2 MS+1.5 mg/L NAA,and the rooting rate reached 81.60%.After 11~15 days of rooting culture and 7~10 days of seedling training in plastic shed,transplanted to a nutrition bowl,the survival rate of transplanting could reach over 90%.
