摘 要:目的明确孕鼠妊娠期糖尿病(GDM)对胎鼠原代培养的肺泡Ⅱ型上皮细胞(AECⅡ)形态、功能的影响及意义。方法SD孕鼠分为对照组和GDM组,GDM组孕鼠建立GDM模型。分离、纯化两组胎鼠的AECⅡ细胞。台盼蓝染色判断胎鼠AECⅡ活力;透射电镜观察胎鼠AECⅡ超微结构改变。苏木素-伊红(HE)染色胎肺组织计算AECⅡ和AECⅠ数量及比例;油红O染色检测胎鼠AECⅡ脂质的变化;免疫荧光检测胎鼠AECⅡ中肺泡表面活性物质相关蛋白C(SP-C,AECⅡ标记物)的表达变化;免疫细胞化学检查胎鼠AECⅡ肺泡表面活性物质相关蛋白A(SP-A)的表达变化。结果原代培养可得到高产量、高纯度的AECⅡ。透射电镜下可见GDM组胎鼠AECⅡ中板层小体数较对照组胎鼠明显减少,细胞表面的微绒毛明显减少。GDM组胎鼠AECⅡ和AECⅠ细胞数量较对照组胎鼠均降低。GDM组胎鼠AECⅡ中脂滴数较对照组胎鼠明显减少。GDM组胎鼠原代培养的AECⅡ中SP-A表达低于对照组胎鼠(P<0.001)。结论GDM时,AECⅡ细胞形态的改变,功能的抑制是新生儿容易出现呼吸窘迫综合征的重要原因。Objective To investigate the effect and significance of gestational diabetes mellitus(GDM)on the morphology and function of primary culture alveolar epithelialⅡ(AECⅡ)cells.Methods The pregnant Sprague-Dawley rats were divided into the control group and the GDM group.The GDM model was established in the GDM group.AECⅡcells were isolated and purified from fetal lungs.The viability of cells was determined by trypan blue staining.Ultrastructure of AECⅡcells were identified by transmission electron microscopy.The total number and proportion of AECⅡand AECⅠcells were calculated in fetal lung fragments stained by HE staining.Lipid droplets in AECⅡcells were observed by oil red O staining.The expression of surface protein C(SP-C),the molecular maker of ACEⅡwas measured by immunofluorescence and protein A(SP-A)was measured by immunohistochemisty.Results High-yield,high-purity of ACEⅡcould be obtained by primary culture.Under transmission electron microscope,the number of lamellar bodies and microvilli on the surface of AECⅡcells in the fetal lung of the GDM group were significantly decreased when compared with the fetal lung of the control group.The proportion of AECⅡcells and AECⅠcells in the fetal lung of the GDM group was lower than that in the fetal lung of the control group,and the number of lipid droplets of AECⅡcells in the fetal lung of the GDM group was significantly decreased when compared with the fetal lung of the control group.The expression of SP-A in AECⅡcells in the fetal lung of the GDM group was lower than that of the control group(P<0.001).Conclusion The morphology and function of AECⅡcells in patients with GDM are inhibited,which may lead to neonatal respiratory distress syndrome.
