摘　　要：采用生物信息分析学方法,通过UniProt数据库检索人、鼠、兔、犬和猴的白介素-12(IL-12)氨基酸序列,运用ClustalW及Espript软件进行两序列比对,运用Blat数据库进行多序列比对,分析了人的IL-12氨基酸序列与其它种属的差异。结果表明,人的IL-12α亚基及IL-12β亚基氨基酸序列与各种属的同源性大小顺序均为猴源>犬源>兔源>鼠源,说明人的IL-12氨基酸序列与恒河猴、食蟹猴的一致性较高,与其它种属的一致性普遍较低,因此存在种属特异性。该研究为IL-12产品的非临床研究和评价中的动物种属选择提供了参考,可指导新药产品的注册申报。We retrieved the amino acid sequences of interleukin-12(IL-12)from human,mouse,rabbit,dog,and monkey from UniProt database by a bioinformatics analysis method.Moreover,we analyzed the differences in IL-12 amino acid sequences of human and other species through pairwise sequence alignment by ClustalW and Espript software and multiple sequence alignment by Blat database.The results show that the order of homology of IL-12αand IL-12βsubunit amino acid sequences between human and other species is monkey>dog>rabbit>mouse,indicating that the amino acid sequences of human IL-12 are in high consistency with rhenus monkey and crab-eating macaque,but in low consistency with other species,thus there is species specificity.This study provides a reference for the selection of animal species in the non-clinical research and evaluation of IL-12 products,and can guide the registration and application of new drug products.
