摘　　要：目的探讨肾穿刺免疫荧光的方法改良。方法对肾穿刺活检标本经冰冻切片机切片,分别用经典方法和改良方法做IgG、IgM、IgA、C1q、C3、Fibrinogen、Ⅳ型胶原α3链、α5链免疫荧光染色。结果冰冻切片完整、没有冰晶,肾小球数量满意,荧光标记定位准确,可见团块状、线状或颗粒状的绿色荧光,且改良法比经典法更方便、快捷、背景干净,结果可靠。结论该改良方法具有方便、快捷、荧光染色结果稳定可靠的特点,适合推广应用。Objective To discuss the improvement of immunofluorescence of renal puncture.Methods Renal biopsy specimens from Children's Hospital of Soochow University were sectioned with frozen microtome,immunofluorescence staining of IgG,IgM,IgA,C1q,C3,Fibrinogen,typeⅣcollagenα3 chain,α5 chain using classical and modified methods,respectively.Result Frozen sections complete without ice crystals,satisfied with the number of glomeruli,accurate fluorescent marker positioning,visible clump,linear or granular green fluorescence,and the improved method is more convenient,faster,and has a cleaner background than the classic method,with reliable results.Conclusion The improved method has the characteristics of convenience,fastness,stable and reliable fluorescent staining results,and is suitable for popularization and application.
