摘  要:目的:观察预电针对D-半乳糖诱导的阿尔茨海默病样病理大鼠早期病理及认知功能的影响。方法:将SD雄性大鼠随机分为正常组、模型组和预电针组,模型组及预电针组大鼠予D-半乳糖连续腹腔注射7周(120 mg·kg-1·d-1)。预电针组大鼠自造模第1天起予电针百会、肾俞治疗,频率50 Hz,电流1 mA,留针20 min,每日1次,共治疗7周。正常组、模型组大鼠在预电针组干预时进行相同的抓取捆绑,1次/d,20 min/次,共计7周。造模及治疗结束后行Morris水迷宫实验评估各组大鼠学习记忆能力。采用透射电镜观察各组大鼠海马CA1区突触后致密带厚度和突触间隙宽度,采用高尔基染色观察各组大鼠海马CA1区树突棘密度,采用免疫组化检测各组大鼠中缝背核区PHF-1水平,采用免疫荧光检测各组大鼠中缝背核区GSK-3β水平。结果:与正常组比较,模型组大鼠水迷宫实验逃避潜伏期显著延长,差异具有统计学意义(P<0.01),目标象限探索时间显著缩短,差异具有统计学意义(P<0.01),出现学习记忆能力损伤;模型组大鼠海马CA1区突触后致密带厚度显著减小,差异具有统计学意义(P<0.01),突触间隙宽度与正常组比较差异无统计学意义(P>0.05),树突棘密度显著降低,差异具有统计学意义(P<0.01),且中缝背核区PHF-1、GSK-3β表达水平显著升高,差异具有统计学意义(P<0.01)。与模型组比较,预电针组大鼠水迷宫实验逃避潜伏期显著缩短,差异具有统计学意义(P<0.01),目标象限探索时间显著延长,差异具有统计学意义(P<0.01),海马CA1区突触后致密带厚度显著升高,差异具有统计学意义(P<0.01),树突棘密度显著升高,差异具有统计学意义(P<0.01),中缝背核区PHF-1、GSK-3β表达水平显著下降,差异具有统计学意义(P<0.01或P<0.01)。结论:预电针可改善阿尔茨海默病样病理大鼠突触结构可塑性损伤及中缝背核区神经原纤维缠结病理沉积,从而改善认知障Objective:To observe the effects of preventive electro-acupuncture on the early pathology and cognitive function in D-galactose induced Alzheimer’s disease(AD)-like rats.Methods:SD male rats were randomly divided into the normal group,the model group and the preventive electro-acupuncture group.AD-like model was established by intraperitoneal injection with D-galactose(120 mg/kg·day)for 7 weeks.At the beginning of modeling,the rats in the preventive electro-acupuncture group were intervened by electro-needling Shenshu(BL23)and Baihui(GV20).After 7 weeks of the intervention,the learning and memorizing ability was assessed by Morris water maze test in the groups.Transmission electron microscopy was used to observe the width of synaptic clefts and the postsynaptic density in hippocampal CA1 area.Golgi staining was performed to observe the dendritic spine density in hippocampal CA1 area.Immunohistochemistry was adopted to detect the PHF-1 expression in dorsal raphe nucleus(DRN)area.Immunofluorescence was applied to detect the expression of GSK-3βin DRN area.Results:The escape latency was significantly prolonged(P<0.01),and the time spent in the target quadrant was significantly shortened in the model group compared to those in the normal group(P<0.01).Compared to that in the normal group,the postsynaptic density was significantly reduced in CA1 area(P<0.01),and the dendritic spine density was significantly decreased in the model group(P<0.01);however,there was no statistical difference in the width of synaptic clefts between the two groups(P>0.05).The expression levels of PHF-1 and GSK-3βin DRN were significantly increased in the model group compared to those in normal group(P<0.01).Compared to that in the model group,the escape latency was significantly shortened(P<0.01),the time spent in the target quadrant was significantly prolonged(P<0.01),the postsynaptic density of CA1 area was significantly increased(P<0.01),the dendritic spine density was significantly also increased(P<0.01),and the expression levels of
