摘  要:目的:探讨头穴丛刺对阿尔茨海默病模型大鼠学习记忆能力及海马区β-APP表达的影响。方法:将SPF级雄性Wistar大鼠随机分为正常组、假手术组、模型组和头穴丛刺组,每组12只。模型组、头穴丛刺组采用双侧海马内注射Aβ1-42诱导AD模型。造模成功后第7天开始干预,头穴丛刺组选取百会穴及百会穴左侧旁开1 mm处、百会穴右侧旁开1 mm进行针刺干预,1次/d,留针30 min,连续干预14 d。应用Morris水迷宫于治疗前、治疗后进行大鼠行为学测试;干预后,采用免疫组化法检测大鼠海马区β-APP蛋白表达;干预后,采用实时荧光定量PCR法检测大鼠海马区β-APP mRNA表达。结果:治疗前和干预14 d后,与假手术组比较,模型组大鼠潜伏期均显著延长(P<0.01),穿台次数均显著减少(P<0.01);干预14 d后,与模型组比较,头穴丛刺组大鼠潜伏期明显缩短(P<0.01),穿台次数明显增加(P<0.01)。与假手术组比较,模型组大鼠海马区β-APP蛋白表达和mRNA表达均显著升高(P<0.01);与模型组比较,头穴丛刺组大鼠海马区β-APP蛋白表达和mRNA表达均明显降低(P<0.01)。结论:头穴丛刺针刺法能有效改善AD大鼠学习记忆能力,其机制可能与调控海马区β-APP蛋白表达有关。Objective:To investigate the effect of scalp clustering acupuncture on learning and memory ability and hippocampal amyloid precursor protein(β-APP)expression in model rats with Alzheimer's disease(AD).Methods:SPF grade male Wistar rats were randomly divided into the normal group,the sham operation group,the model group and the scalp clustering acupuncture group,with 12 rats in each group.AD model was induced by bilateral intra-hippocampal injection of Aβ1-42.The intervention began on 7 d after successful modeling,the points chosen in the scalp clustering acupuncture group were Baihui(DU20),points 1mm left and right lateral to DU20,once per day for 14 continuous days.Morris water maze was used to test rat behavior before and after the treatment;after the intervention,the expression of hippocampalβ-APP protein was detected by immunohistochemical method,and the expression of hippocampalβ-APP mRNA was detected by real-time fluorescence quantitative PCR.Results:Before and after 14 d of the intervention,the incubation period was significantly prolonged in the model group(P<0.01),and the number of crossing platform was significantly reduced in the model group compared to those in the sham operation group(P<0.01);the incubation period was significantly shortened(P<0.01),and the number of crossing platform was significantly increased in the scalp clustering acupuncture group after 14 d of the intervention compared to those in the model group(P<0.01).The protein and mRNA expressions ofβ-APP in the hippocampus were significantly increased in the model group compared to those in the sham operation group(P<0.01);which were significantly decreased after the intervention of scalp clustering acupuncture(P<0.01).Conclusion:Scalp clustering acupuncture can effectively improve learning and memory ability of AD rats,which mechanism may be related to the regulation ofβ-APP protein expression in hippocampus.
