摘  要:【目的】为明确楚雄市紫溪山华山松种子园内各种源无性系的遗传结构及其亲缘关系,对该种子园内6个种源的97个无性系进行SSR遗传多样性分析。【方法】利用改良CTAB法,提取97个华山松无性系DNA,用前期筛选出的26对高多态性引物对所有DNA样品进行SSR-PCR扩增,并统计其多态性条带信息,最后用POPGEN 1.32、NTSYS 2.10和Structure 2.3.1软件对其遗传多样性进行分析。【结果】(1)在群体水平上,6个种源的N_(a)范围为3.7308~4.8077,均值为4.2371,N_(e)为2.9011,H为0.6376,I为1.1865,表明华山松各种源均具有较高的遗传多样性。(2)聚类分析表明:地理距离较近的种源其遗传差异并不一定近,UPGMA聚类图并未将地理位置较近的种源归为一类。(3)华山松的基因流N_(m)均值为4.1503,说明华山松种源间的遗传交流较多。【结论】华山松无性系种源间遗传变异不高,其主要遗传变异存在于种源内,研究结果可为利用该种子园内的不同无性系进行华山松遗传改良提供理论依据及实践指导。[Purpose]In order to illuminate the genetic structure and genetic relationship of various provenance Pinus armandii clones from seed orchard of Zixi Mountain in Chuxiong City,SSR genetic diversity analysis was performed on 97 clones from six provenances of the seed orchard.[Method]The modified CTAB method was used to extract the DNA of 97 P.armandii colones,and then 26 pairs of high-polymorphism primers screened in the previous stage were used for SSRPCR amplification of all DNA samples,and their polymorphism band information was counted.Finally,POPGEN 1.32,NTSYS 2.10 and Structure 2.3.1 software were used to analyze their genetic diversity.[Result]1)At the population level,the range of N_(a) in six provenances was from 3.7308 to 4.8077,with an average of 4.2371,while N_(e) was 2.9011,H was 0.6376,I was 1.1865.It showed that P.armandii of different provenance all had high genetic diversity.2)The cluster analysis showed that the genetic differences of provenances with close geographical distance were not necessarily close,and the UPGMA cluster diagram did not classify the provenances with close geographical location into one group.3)The average N_(m) value of P.armandii gene flow was 4.1503,indicating that there was more genetic exchange among P.armandii provenances.[Conclusion]The genetic variation among provenances of P.armandii clones was not high,and the main genetic variation was within the provenance.The results of this study could provided theoretical basis and practical guidance for the genetic improvement of P.armandii in seed orchard by using different clones.
