摘  要:目的建立同时测定泌炎宁颗粒中绿原酸、蒙花苷含量的反相高效液相色谱法。方法色谱柱为ACE Excel C_(18)柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸水溶液(梯度洗脱),流速为1.0 m L/min,检测波长为334 nm,柱温为30℃,进样量为10μL。结果绿原酸、蒙花苷质量浓度分别在11.33~135.91μg/m L(r=0.9992,n=5)和5.48~65.75μg/m L(r=0.9990,n=5)范围内与峰面积线性关系良好;精密度、稳定性、重复性试验的RSD均小于2.0%;平均加样回收率分别为99.44%和99.31%,RSD分别为1.87%和5.34%(n=9)。结论该方法操作简便,重复性好,准确度高,可用于同时测定泌炎宁颗粒中绿原酸、蒙花苷的含量。Objective To establish a reverse phase-high performance liquid chromatography(RP-HPLC)method for the simultaneous determination of chlorogenic acid and buddleoside in Miyanning Granules.Methods The chromatographic column was ACE Excel C_(18) column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.1%phosphoric acid aqueous solution(gradient elution),the flow rate was 1.0 m L/min,the detection wavelength was 334 nm,the column temperature was 30℃,and the injection volume was 10μL.Results Chlorogenic acid and buddleoside showed a good linear relationship in the ranges of 11.33-135.91μg/m L(r=0.9992,n=5)and 5.48-65.75μg/m L(r=0.9990,n=5),respectively.The RSDs of precision,stability and repeatability tests were less than 2.0%.The average recoveries of chlorogenic acid and buddleoside were 99.44%and 99.31%with RSDs of 1.87%and 5.34%(n=9),respectively.Conclusion The method is simple,reproducible and accurate,which can be used for the simultaneous determination of chlorogenic acid and buddleoside in Miyanning Granules.
