摘  要:为研究miR398和miR408对UV-B辐射下拟南芥幼苗的影响,测定了叶绿素荧光参数、抗氧化系统SOD酶活性、GSH含量和MDA含量等指标.结果表明:UV-B辐射下,过表达miR398拟南芥与过表达miR408拟南芥相比野生型植株的叶绿素荧光动力学参数F_(v)/V_(m)、qP降低、qN升高,影响植物的光合作用过程,降低光合效率;SOD酶活性降低、GSH含量减少,MDA含量上升,活性氧积累,影响植物抵抗辐射逆境胁迫,损害植株生长发育过程.说明过表达miR398和过表达miR408拟南芥对UV-B辐射胁迫的抗性降低,光合作用和抗氧化系统均受到影响,miR398与miR408对拟南芥响应UV-B胁迫下起负调控作用.To study the regulation of miR 398 and miR 408 on the photosynthetic function and antioxidantsystem of Arabidopsis thaliana leaves under UV-B radiation.The chlorophyll fluorescence parameters related tophotosynthesis and antioxidant system related indicators including SOD enzyme activity,GSH and MDA contentwere detected.The results showed that the chlorophyll fluorescence kinetic parameters F_(v)/F_(m),qP,and qN in-creased in wild-type plants compared to over-expressed miR 398Arabidopsis and overexpressed Arabidopsis underUV-B radiation.The photosynthesis process of plants reduces the photosynthetic efficiency;the activities of an-tioxidant enzymes SOD activity and GSH content decreased,the content of oxidizing substances MDA rised,andthe accumulation of active oxygen affects the resistance of plants to radiation stress and damages the growth anddevelopment of plants.It showed that resistance to UV-B radiation of over-expressed miR 398 and miR 408 Arabi-dopsis stress decreased,photosynthesis and antioxidant system were affected,miR 398 and miR 408 Arabidopsisunder UV-B play an important negative regulatory role.
