摘 要:为了有效利用栎属植物资源,利用SSR标记技术对栎亚属4个树种24个单株进行亲缘关系分析。结果表明:从44对SSR引物中筛选出9对多态性引物,PCR结果显示,这9对引物在24份材料中共检测到62个等位基因变异,每对引物检测到的等位基因变异范围为4—12个,平均每对引物检测到6.8个等位基因变异。各位点的平均有效等位基因数为4.9633,Nei多样性指数分布范围在0.6285—0.8915,平均值为0.7586,Shannon信息指数为1.1843—2.3367。UPGMA法聚类分析结果显示,24份种质资源间遗传相似系数的平均值为0.7008,变化范围为0.4928—0.9710。栎亚属4个树种分为两大类,中国江苏收集的麻栎单独属于一大类,美国收集的3种树种属于一大类,其中落叶型的沼生栎和常绿型的弗吉尼亚栎为一类,常绿型的墨西哥白橡单独属于一类。研究可为栎属植物引种资源的管理及评价提供重要依据。In order to make effective use of the Quercus resources,24 individuals of four Quercus species were analyzed by SSR marker,and 9 pairs of polymorphic primers were selected from 44 SSR primers for amplification.A total of 62 alleles were detected in 24 materials,ranging from 4 to 12,with an average of 6.8 alleles per pair of primers.The average value of effective allelic variation of each site was 4.9633.The Nei diversity index ranged from 0.6285 to 0.8915,with an average of 0.7586.The Shannon’s information index ranged from 1.1843 to 2.3367.The DNA fingerprints of the 24 individuals of the 4 tree species were obtained by UPGMA method based on the genetic similarity coefficient matrix among individuals,the results indicated that the average genetic similarity coefficient among 24 germplasm resources was 0.7008,and the variation ranged from 0.4928 to 0.9710.The clustering results showed that all the resources were divided into two categories,the oak trees collected from Jiangsu,China belonged to a category,and the three species collected from the United States of America belonged to a category.Among them,deciduous marsh oak and evergreen Virginia Quercus were clustered into a class,while the evergreen Mexican white oak was clustered into a class.The results can provide important basis for the management and evaluation of introduced Quercus resources.
