摘  要:目的:基于Leptin/c⁃Myc/PGC⁃1信号通路调控的能量代谢途径探索黄连⁃吴茱萸配伍抗胃癌的分子生物学机制。方法:以不同浓度(0、1、5、10 mg/mL)黄连⁃吴茱萸提取物干预胃癌SGC7901细胞后,采用MTT法检测其对细胞增殖活性的影响;相差显微镜检查细胞形态;用流式细胞术(FCM)检测对细胞凋亡后周期分布的影响;试剂盒检测细胞中线粒体膜电位和ATP含量的变化;采用荧光定量聚合酶链式反应(RT⁃PCR)和Western blot检测细胞中Leptin/c⁃Myc/PGC⁃1的表达量。结果:与空白对照组比较,黄连⁃吴茱萸5、10 mg/mL组SGC7901细胞活力显著降低,G_(0)/G_(1)期细胞比例显著升高,S期细胞比例显著降低,细胞中JC⁃1单体形式显著升高,ATP含量显著降低;细胞中c⁃Myc、PGC⁃1 mRNA及蛋白表达显著升高,Leptin mRNA及蛋白表达显著降低。结论:黄连⁃吴茱萸配伍可显著抑制SGC⁃7901细胞增殖,升高细胞中JC⁃1单体形式,降低ATP含量,提示黄连⁃吴茱萸配伍在胃癌细胞增殖中具有抑制作用,这可能与调节Leptin/c⁃Myc/PGC⁃1通路有关。Objective:To explore the molecular biological mechanism of the combination of Coptidis Rhizoma⁃Euodiae Fructus against gastric cancer based on the energy metabolism pathway regulated by Leptin/c⁃Myc/PGC⁃1 signaling pathway.Methods:After different concentrations(0,1,5,10 mg/mL)of Coptidis Rhizoma⁃Euodiae Fructus extracts were used to intervene SGC7901 cells in gastric cancer,MTT assay was used to detect its effect on cell proliferation activity,cell morphology was examined by phase contrast microscopy,flow cytometry(FCM)was used to detect the effect on the cycle distribution after apoptosis,the changes of mitochondrial membrane potential and ATP content in cells were detected by the kit.Leptin/c⁃Myc/PGC⁃1 expression in cells was detected by fluorescence quantitative polymerase chain reaction(RT⁃PCR)and Western blot.Results:Compared with the normal control group,the activity of SGC7901 cells in the 5,10 mg/mL Coptidis Rhizoma⁃Euodiae Fructus groups was significantly decreased,the proportion of cells in G_(0_/G_(1) period was significantly increased,the proportion of cells in S period was significantly decreased,the monocyte form of JC⁃1 in the cells was significantly increased and the content of ATP was significantly decreased;The expressions of c⁃Myc,PGC⁃1 mRNA and proteins in cells were significantly increased,while the expressions of Leptin mRNA and protein were significantly decreased.Conclusion:The combination of Coptidis Rhizoma and Euodiae Fructus can significantly inhibit the proliferation of SGC⁃7901 cells,increase the monocyte form of JC⁃1 and reduce the content of ATP,suggesting that the combination of Coptidis Rhizoma and Euodiae Fructus can inhibit the proliferation of gastric cancer cells,which may be related to the regulation of Leptin/c⁃Myc/PGC⁃1 signaling pathway.
