摘 要:目的:研究北柴胡Bupleurum chinense DC.地上部分的黄酮类化学成分。方法:综合运用硅胶柱色谱、ODS柱色谱、Sephadex LH⁃20凝胶柱色谱以及半制备型高效液相色谱等现代色谱分离技术对北柴胡地上部分的黄酮类化学成分进行系统研究。利用NMR和HR⁃MS等波谱学技术并结合参考文献对分离得到的化合物进行结构鉴定。结果:从北柴胡地上部分的70%乙醇提取物中分离得到了12个黄酮类化合物,分别鉴定为:木犀草素(1)、异鼠李素(2)、异鼠李素⁃7⁃O⁃α⁃L⁃鼠李糖苷(3)、苜蓿素⁃7⁃O⁃β⁃D⁃吡喃葡萄糖苷(4)、vittariflavone(5)、tricin⁃4′⁃O⁃[threo⁃β⁃guaiacyl⁃(9″⁃O⁃acetyl)⁃glyceryl]ether(6)、tricin⁃4′⁃O⁃(threo⁃β⁃guaiacylglyceryl)ether⁃7⁃O⁃β⁃D⁃glucopyranose(7)、tricin⁃4′⁃O⁃(threo⁃β⁃guaiacylglyceryl)ether 7″⁃O⁃β⁃D⁃glucopyranose(8)、salcolin B(9)、槲皮素⁃3⁃O⁃β⁃D⁃吡喃葡萄糖醛酸乙酯(10)、兔毛蒿素(11)、(-)⁃(5S,6S)⁃5,6⁃dihydro⁃3,8,10⁃trihydroxy⁃5⁃(4⁃hydroxy⁃3⁃methoxy⁃phenyl)⁃6⁃hydroxymethyl⁃2,4⁃dimet⁃hoxy⁃7H⁃benzo[c]xanthen⁃7⁃one(12)。结论:其中,化合物9为首次从柴胡属植物中分离得到,化合物4~8和10~12为首次从伞形科中分离得到。Ojective:To study the flavonoids from the aerial parts of Bupleurum chinense.Methods:The flavonoids from the aerial parts of Bupleurum chinense were isolated and purified by modern chromatographic separation techniques,such as silica gel column chromatography,ODS column chromatography,Sephadex LH⁃20 gel column chromatography and semi⁃preparative HPLC.Their struc⁃tures were identified by NMR,HR⁃MS and other spectroscopic techniques,as well as comparisons with the data reported in the litera⁃tures.Results:Twelve flavonoids were isolated from the 70%ethanol extract of the aerial parts of Bupleurum chinense,and identified as luteolin(1),isorhamnetin(2),isorhamnetin⁃7⁃O⁃α⁃L⁃rhamnopyranoside(3),tricin⁃7⁃O⁃β⁃D⁃glucopyranose(4),vittariflavone(5),tri⁃cin⁃4′⁃O⁃[threo⁃β⁃guaiacyl⁃(9″⁃O⁃acetyl)⁃glyceryl]ether(6),tricin⁃4′⁃O⁃(threo⁃β⁃guaiacylglyceryl)ether⁃7⁃O⁃β⁃D⁃glucopyranose(7),tricin⁃4′⁃O⁃(threo⁃β⁃guaiacylglyceryl)ether⁃7″⁃O⁃β⁃D⁃glucopyranose(8),salcolin B(9),quercetin⁃3⁃O⁃β⁃D⁃ethyl glucuronate(10),fili⁃folin(11),(-)⁃(5S,6S)⁃5,6⁃dihydro⁃3,8,10⁃trihydroxy⁃5⁃(4⁃hydroxy⁃3⁃methoxy⁃phenyl)⁃6⁃hydroxymethyl⁃2,4⁃dimethoxy⁃7H⁃benzo[c]xanthen⁃7⁃one(12),respectively.Conclusion:Among them,compound 9 is isolated from Bupleurum genus for the first time,com⁃pounds 4 to 8 and 10 to 12 are isolated from Umbelliferae for the first time.
