救必应HPLC指纹图谱及体外抗病毒活性的谱效关系研究

摘　　要：目的:建立救必应的HPLC指纹图谱,研究其与体外抗病毒活性的关系。方法:采用HPLC法建立救必应的指纹图谱;利用“中药色谱指纹图谱相似度评价系统(2012版)”进行相似度评价;使用抗流感病毒神经氨酸酶(NA)效价试剂盒检测救必应提取物对NA的体外抑制活性;利用灰色关联度分析和偏最小二乘回归分析法研究二者的关系。结果:建立了含有25个共有峰的12批救必应HPLC指纹图谱,通过对照品比对,指认了其中4个成分。谱效关系表明8、12、14、18、20和24号色谱峰与抗NA活性关联较大。结论:救必应的体外抗病毒活性是多种成分共同作用的结果,该研究为救必应的质量控制提供了参考。Objective:To investigate the HPLC fingerprint of Ilicis Rotundae CoRte,and to study its relationship with antiviral ac⁃tivity in vitro.Methods:HPLC method was used to establish fingerprint of Ilicis Rotundae CoRte.The similarity evaluation was carried out by using the“Similarity evaluation system for TCM chromatographic fingerprints(2012 edition)”.The in vitro inhibitory activity of the extracts was detected with the anti⁃influenza virus neuraminidase(NA)titer kit.Grey relational degree analysis and partial least squares regression analysis were used to study the relationship between them.Results:HPLC fingerprint of 12 batches of Ilicis Rotundae CoRte containing 25 common peaks was established,and four components were identified by comparison of the control substances.The spectrum⁃effect relationship results showed that 8,12,14,18,20 and 24 peaks were significantly correlated with the activity of anti⁃NA.Conclusion:The antiviral activity in vitro of Ilicis Rotundae CoRte is the result of the combined action of many components.This study provides a reference for the quality control of Ilicis Rotundae CoRte.
