摘  要:目的:阐明葛根和粉葛治疗高血压的药效区别及机制。方法:采用腹腔注射L⁃NAME的方法制作高血压大鼠模型,将大鼠分为9组:正常对照组、模型组、卡托普利组、葛根(低、中、高剂量)组、粉葛(低、中、高剂量)组。造模成功后给药组分别给予相应药物4 w,每周测定大鼠血压,4 w后腹主动脉取血并摘取心脏和胸主动脉组织。试剂盒测定血浆NO、eNOS、LDH、SOD、ANP含量,称量质量计算心脏指数,HE染色观察心脏和胸主动脉组织病理变化。结果:与模型组比较,葛根各剂量组大鼠血压、心脏指数及血浆LDH含量显著降低,血浆NO、eNOS、SOD含量显著升高,葛根中、高剂量组血浆ANP含量显著降低(P<0.05~P<0.001),而粉葛对上述指标无明显影响。组织病理学检查显示,葛根改善了大鼠心肌肥大、心肌纤维化和水肿及血管内皮细胞肿胀有破裂和中膜平滑肌细胞增生,而粉葛对心脏和血管组织无明显改善。结论:葛根对L⁃NAME诱导的高血压大鼠具有治疗作用,而粉葛对高血压大鼠无明显疗效。葛根治疗高血压的机制和提高eNOS和SOD活性及调节ANP分泌有关。Objective:To elucidate the pharmacodynamic differences and mechanism between Puerariae Lobatae Radix and Puerar⁃iae Thomsonii Radix in the treatment of hypertension.Methods:The hypertensive rat model was prepared by intraperitoneal injection of L⁃NAME.The rats were divided into 9 groups,including normal control group,model group,captopril group,Puerariae Lobatae Radix(low,medium and high doses)group and Puerariae Thomsonii Radix(low,medium and high doses)group.After successful modeling,the drug gruop was given the corresponding drug for 4 weeks,and the blood pressure was measured weekly.Blood was taken from the ab⁃dominal aorta after 4 weeks and hearts and thoracic aortic tissues were extracted.The contents of NO,eNOS,LDH,SOD and ANP in plasma were measured with the kit,the heart index was calculated by weighing the mass.The pathological changes of hearts and thoracic aorta tissues were observed by HE staining.Results:Compared with the model group,Puerariae Lobatae Radix gruops with each doase could significantly reduce blood pressure,heart index and the contents of LDH and ANP in plasma,the contents of NO,eNOS and SOD in plasma were significantly increased,the content of ANP in plasma of the medium and high doses of Puerariae Lobatae Radix was sig⁃nificantly decreased(P<0.05~P<0.001),while Puerariae Thomsonii Radix had no significant effects on the above inde⁃xes.Histopathological examination showed that Puerariae Lobatae Radix could improve myocardial hypertrophy,myocardial fibrosis and edema,the swelling of vascular endothelial cells with rupture and proliferation of mediummembrane smooth muscle cells in rats,while Puerariae Thomsonii Radix had no significant improvement in heart and vascular tissues.Conclusion:Puerariae Lobatae Radix has thera⁃peutic effect on L⁃NAME induced hypertension in rats,but Puerariae Thomsonii Radix has no significantly effect on hypertensive rats.The mechanism of Puerariae Lobatae Radix in the treatment of hypertension is related to the improvement of eNOS and SOD acti
