摘  要:目的探讨水飞蓟素通过Caspase-3表达诱导子宫内膜癌细胞凋亡的药理机制。方法选取2019年1月—2020年4月江西省妇幼保健院收治的60例子宫内膜癌患者,取患者的人子宫内膜癌Ishikawa细胞株,设置对照组(不加任何药物)、观察组(加入水飞蓟素),应用四唑盐(MTT)比色法检验水飞蓟素对人子宫内膜癌Ishikawa细胞增殖影响,使用流式细胞术检测细胞凋亡率,使用免疫组化法检验Caspase-3表达水平。结果观察组人子宫内膜癌Ishikawa细胞吸光度低于对照组(P<0.001);观察组人子宫内膜癌Ishikawa细胞凋亡率高于对照组(P<0.001);观察组中人子宫内膜癌Ishikawa细胞Caspase-3表达水平高于对照组(P<0.001)。结论水飞蓟素可通过上调细胞Caspase-3的表达,诱导子宫内膜癌细胞凋亡。Objective To investigate the pharmacological mechanism of silymarin induced apoptosis of endometrial cancer cells by Caspase-3 expression.Methods 60 patients with endometrial cancer were selected as the research object from January 2019 to April 2020 in Jiangxi Maternal and Child Health Hospital.The human endometrial cancer cell lines were obtained,and the control group(without any drugs)and silymarin group(with silymarin)were set up.The effect of silymarin on endometrial cancer cell activity was tested by MTT colorimetry,and flow cytometry was used to detect The expression of caspase-3 protein was detected by immunohistochemistry.Results The absorbance of silymarin group was lower than that of control group(P<0.001).Flow cytometry showed that the apoptosis rate of Ishikawa cells in the silymarin group was higher than that in the control group(P<0.001).The expression level of Caspase-3 protein in silymarin group was higher than that in control group(P<0.001).Conclusion Silymarin can induce apoptosis of endometrial cancer cells by upregulating the expression of Caspase-3.
