摘  要:目前关于鸭(Anas platyrhynchos)中肌细胞增强因子2C基因(myocyte enhancer factor 2C, MEF2C)剪切体的表达尚未开展深入研究。为了解MEF2C基因2个剪切体在组织中的表达差异,本研究运用RTPCR技术克隆兴义鸭MEF2C基因的全长CDS;运用qPCR技术,检测MEF2C基因在兴义鸭(公和母) 5个不同发育时期的组织表达。结果显示,克隆获得鸭MEF2C基因2个剪切体MEF2C-a (GenBank No.MZ497418)和MEF2C-b (GenBank No. MZ497419);2个剪切体所编码的氨基酸序列与原鸽(Columba livia)、鹌鹑(Coturnix japonica)、原鸡(Gallus gallus)等物种的同源性高达95%以上,说明MEF2C基因在进化过程中具有较高的保守性。qPCR结果显示,MEF2C-a和MEF2C-b在公鸭和母鸭组织中均有表达,在脑、心肌、腿肌和胸肌组织中高表达;2个剪切体在母鸭和公鸭组织中的表达规律不同,在公鸭多数组织中MEF2C-a表达量高于MEF2C-b,在母鸭中则相反;2个剪切体在腿肌和胸肌中的表达不随日龄的增加而升高。本研究结果为进一步探讨MEF2C基因不同剪切体在鸭肌肉发育过程中的作用机制提供基础资料。At present, the expression of splice variants of myocyte enhancer factor 2 C gene(MEF2 C) in ducks(Anas platyrhynchos) has not been deeply studied. In order to understand the expression difference of MEF2 C variants in tissues, RT-PCR was used to clone the full-length CDS of MEF2 C gene in Xingyi duck.The results showed that 2 splice variants of MEF2 C-a(GenBank No. MZ497418) and MEF2 C-b(GenBank No. MZ497419) were successfully cloned, and the encoded amino acid sequences shared more than 95%homology with pigeon(Columba livia), quail(Coturnix japonica) and chicken(Gallus gallus), which indicated that MEF2 C gene was highly conserved in the evolution process. The results of qPCR showed that MEF2 C-a and MEF2 C-b were expressed in both male and female ducks, and highly expressed in brain, myocardium, leg muscle and chest muscle. The expression patterns of the 2 splice variants in female and male ducks were different. The expression level of MEF2 C-a in most tissues of male ducks was higher than that of MEF2 C-b,the opposite in female ducks. The expression of 2 splice variants in leg muscles and chest muscles did not increase with increasing age. The present results provide basic materials for further study on the mechanism of MEF2 C splice variants in duck muscle development.
