摘  要:目的:本研究分析比较霍山石斛、细茎石斛和铁皮石斛的遗传关系。方法:以黄石斛为参考基因组,用测序基因分型技术获得单链核苷酸多态性,根据单链核苷酸多态性差异,分别用Admixture软件分析3种石斛的居群遗传结构,用TreeBeST软件构建邻接树,用GCTA软件做主成分分析,用GenAlEx软件计算遗传距离。结果:3种石斛平均每个样本产生的干净片段(clean reads)和单链核苷酸数目都是霍山石斛>细茎石斛>铁皮石斛;邻接树结果表明铁皮石斛样本聚为一支,霍山石斛样本和细茎石斛样本聚为另一支,且该支的霍山石斛样本与细茎石斛样本又被严格分开为2小支;遗传结构和主成分分析结果与邻接树类似;霍山石斛与细茎石斛间的遗传距离小于霍山石斛与铁皮石斛间的遗传距离。结论:根据3种石斛的核苷酸多态性差异,3种石斛能被明显分开;霍山石斛与细茎石斛间的遗传关系近于霍山石斛与铁皮石斛间的遗传关系。Objective:The genetic relationships among Dendrobium huoshanense,Dendrobium moniliforme and Dendrobium officinale were investigated in the study.Methods:The number of single nucleotide polymorphisms(SNP)were obtained according to genotyping by sequencing(GBS)analysis,using Dendrobium catenatum as the reference genome. The genetic structure was constructed by Admixture software,Neighbor-joining phylogenetic tree(NJ tree)was drawn through TreeBeST,Principal component analysis(PCoA)was built using GCTA. The genetic distance was computed with GenAlEx software,based on the differennce of SNPs diversity in 3 Dendrobium species.Results:Among 3 Dendrobium species,the number of clean reads and SNPs per each specimen ranging from high to low respectively were D. huoshanense,D. moniliforme,and D. officinale. The NJ tree showed that all specimens were grouped into two braches. One branch was composed by all specimens of D. officinale. The other branch was divided into two clusters,one cluster was consisted of all D. huoshanense specimens,while the other cluster was comprised of all D. moniliforme specimens. The results of genetic structure and PCoA were similar with that of NJ tree;Genetic distance between D. huoshanense and D. moniliforme was lower than that between D. huoshanense and D. officinale.Conclusion:The results showed that 3 Dendrobium species were obviously separated based on the difference of SNPs diversity. The genetic relationship between D.huoshanense and D. moniliforme was relatively closer than that between D. huoshanense and D. officinale.
