摘 要:目的:观察咳嗽变异性哮喘(CVA)大鼠肺功能、肺组织病理学变化,探讨紫诃朴喘颗粒对CVA大鼠的干预作用。方法:大鼠随机分为正常组,模型组,醋酸地塞米松组,苏黄止咳胶囊组,紫诃朴喘颗粒高、中、低剂量组,每组12只。采用卵蛋白腹腔注射的方法制备CVA大鼠模型,各组均给予相应药物灌胃,连续14d。检测大鼠气道反应性,观察肺组织病理学变化。结果:与正常组比较,模型组在雾化PBS时大鼠气道阻力显著升高(P<0.05);与模型组比较,紫诃朴喘颗粒各剂量组及醋酸地塞米松组气道阻力显著降低(P<0.05,P<0.01);与正常组比较,在乙酰甲胆碱浓度为3.125、50mg/mL时,模型组大鼠气道阻力显著升高(P<0.05,P<0.01);与模型组比较,在乙酰甲胆碱浓度为50mg/mL时,紫诃朴喘颗粒低剂量组气道阻力显著降低(P<0.05),其他各给药组均有下降趋势。模型组大鼠肺组织支气管管壁出现结构变化,肺间质炎性细胞增多,支气管内分泌物大量增多,部分出现瘀血。紫诃朴喘颗粒低、中剂量组肺泡损伤较轻,结构较完整,间质水肿不明显;气道分泌物明显减少,偶见炎性细胞浸润。结论:紫诃朴喘颗粒可在一定程度上降低CVA大鼠气道阻力,减轻肺泡损伤,减少肺间质炎性细胞浸润,并减少气道分泌物的产生,明显改善肺组织病理学变化。Objective: To observe the changes of lung function and lung histopathology in cough variant asthma(CVA)rats, and to explore the intervention effect of Zihepuchuan Granules on CVA model rats. Methods: Rats were randomly divided into normal control group, model group, dexamethasone acetate group, Suhuang Zhike Capsules group and Zihepuchuan Granules high,middle and low dose groups, 12 rats in each group. The CVA rat model was established by intraperitoneal injection of ovalbumin, and each group was given corresponding drugs by intragastric administration for 14 days. The airway reactivity of rats and observe the pathological changes of lung tissue were detected. Results: Compared with the normal group, airway resistance in the model group increased significantly(P<0.05). Compared with the model group, the airway resistance of Zihe Puchuan Granules in each dose group and dexamethasone acetate group were significantly reduced(P<0.05, P<0.01). Compared with the normal group, the airway resistance of rats in the model group was significantly increased when the concentration of acetylcholine was 3.125 mg/mL and 50 mg/mL(P<0.05,P<0.01). Compared with the model group, when the concentration of methacholine was 50 mg/mL, the airway resistance in the low dose Zihepuchuan Granules group was significantly lower(P<0.05), and there was a downward trend in other administration groups. In the model group, there were structural changes in the bronchial wall of lung tissue, an increase of interstitial inflammatory cells, a large increase of bronchial endocrine secretion, and some congestion. In the low and middle dose Zihepuchuan Granules groups, the alveolar injury was mild, the structure was intact, the interstitial edema was not obvious, the airway secretion was significantly decreased, and inflammatory cell infiltration was occasionally seen. Conclusion: Zihepuchuan Granules can reduce airway resistance, alveolar injury,pulmonary interstitial inflammatory cell infiltration, airway secretion and lung histopathological changes in
