摘 要:目的:探讨益肾降浊冲剂对大鼠尿毒症心肌病心脏Bcl-2、Bax的影响。方法:5/6肾切除术制作尿毒症心肌病模型。SD大鼠随机分为6组:假手术组、模型组、大黄素组及益肾降浊冲剂低、中、高剂量组。益肾降浊冲剂低、中、高剂量组及大黄素组在大鼠术后予益肾降浊冲剂、大黄素灌胃,假手术组和模型组大鼠术后予等量蒸馏水灌胃。免疫组织化学法观察心脏组织中Bcl-2、Bax表达情况;Western blot及RT-qPCR检测心脏组织中Bcl-2、Bax蛋白及mRNA表达情况。结果:与假手术组比较,模型组Bax蛋白及mRNA表达显著上升(P<0.01),Bcl-2蛋白及mRNA表达显著下降(P<0.01);与模型组比较,益肾降浊冲剂中、高剂量组Bax蛋白及mRNA表达显著下降(P<0.05,P<0.01),Bcl-2蛋白及mRNA表达显著上升(P<0.05,P<0.01)。结论:益肾降浊冲剂对尿毒症心肌病心脏的保护作用可能是与增加大鼠心肌组织中Bcl-2的表达,降低其Bax的表达有关。Objective: To explore the effects of Yishen Jiangzhuo Granules on Bcl-2 and Bax in the heart of uremic cardiomyopathy rats. Methods: Uremic cardiomyopathy model was made by 5/6 nephrectomy. SD rats were randomly divided into 6 groups: sham operation group, model group, emodin group, low, medium and high dose Yishen Jiangzhuo Granules groups. Rats in the low, middle and high dose Yishen Jiangzhuo Granules groups and emodin group were given Yishen Jiangzhuo Granules and emodin by gavage after operation, rats in the sham operation group and model group were given distilled water by gavage after operation. Immunohistochemistry was used to observe the expression of Bcl-2 and Bax in heart tissue;Western blot and RTqPCR were used to detect the expression of Bcl-2, Bax protein and mRNA in heart tissue. Results: Compared with sham operation group, the protein and mRNA expression of Bax in the model group was significantly increased(P<0.01), the protein and mRNA expression of Bcl-2 was significantly decreased(P<0.01), and the protein and mRNA expression of Bcl-2 was significantly decreased(P<0.01). Compared with the model group, the protein and mRNA expression of Bax in the middle and high dose Yishen Jiangzhuo Granules groups decreased significantly(P<0.05, P<0.01), and the protein and mRNA expression of Bcl-2 increased significantly(P<0.05, P<0.01). Conclusion: The protective effect of Yishen Jiangzhuo Granules on the heart of uremic cardiomyopathy may be related to the increase of bcl-2 expression and the decrease of Bax expression.
