摘  要:目的制备一种三维有序大孔明胶材料并用于调节难溶性药物非洛地平的释放行为。方法采用模板法制备三维有序大孔明胶材料(three-dimensional ordered macroporous gelatin,TDMG)。采用吸附平衡法将非洛地平(felodipine,FLDP)载入TDMG的三维有序大孔孔道中以制备FLDP-TDMG载药样品。通过扫描电子显微镜(scanning electron microscope,SEM)观察TDMG的结构特征。采用差式扫描量热法(differential scanning calorimetry,DSC)和X射线衍射(X-ray diffraction,XRD)分析FLDP在TDMG大孔孔道中的存在状态。体外溶出实验考察FLDP的药物释放规律。降解实验和膨胀度实验用于评价TDMG性质。结果TDMG具有三维有序大孔结构,载药后,FLDP是以微晶状态存在于TDMG的三维孔道中,载药样品呈现缓释效果。材料具备可降解性,膨胀度实验结果解释了TDMG的调节药物释放速率的机制。结论TDMG能够抑制药物结晶度,调节FLDP的释放速率,具有改善难溶性药物生物利用度的应用前景。Objective To prepare a three-dimensional ordered macroporous gelatin(TDMG),and regulate the release behavior of felodipine(FLDP).Methods The TDMG was prepared by template method.FLDP was loaded into the three-dimensional ordered channels of TDMG by adsorption equilibrium method to prepare drug-loaded samples(FLDP-TDMG).The structural characteristics of TDMG were observed by scanning electron microscope(SEM).Differential scanning calorimetry(DSC)and X-ray diffraction(XRD)were used to analyze the existence of FLDP in the channels of TDMG.The release of FLDP was investigated by in vitro dissolution test.Degradation test and dilatancy test were used to evaluate TDMG properties.Results TDMG had a three-dimensional ordered macroporous structure.After drug loading,FLDP existed in the three-dimensional channels of TDMG in the form of microcrystalline,and the FLDP-TDMG showed sustained release effect.The material was degradable and the dilatancy test results explained the mechanism of regulating the drug release rate of TDMG.Conclusion TDMG can inhibit the drug crystallinity and regulate the release rate of FLDP,which has the application prospect of improving the bioavailability of insoluble drugs.
