摘 要:目的研究守壳聚糖(chitosan,Chi)与胶原(collagen,Col)的复合支架,作为牙周组织工程材料的物理性能和生物安全性,探讨其作为生物材料的可行性。方法采用冷冻干燥法将Chi和Col按不同比例制成混合支架,支架的孔隙率用液体置换法测定,扫描电镜下观察支架的孔径,并进行皮内刺激试验,热原试验等生物材料的安全性试验。结果扫描电镜显示Chi-Col支架具有良好的多孔网格结构,孔径约为100μm。当Chi∶Col为1∶2时,孔隙率和孔径大小最符合组织工程材料的要求且与其他组相比具有显著的差异(P<0.05),不同比例Chi-Col支架材料弹性模量和最大拉伸强度检测结果显示,当Chi∶Col为1∶2时弹性模量和最大拉伸强度与其他两组相比差异有显著性(P<0.05)。在进行急性全身毒性试验的前后,小鼠状态良好。实验侧皮内刺激实验阴性,与各对照侧无明显差异。结论壳聚糖-胶原复合支架具有良好的生物安全性和物理性能,是一种安全的植入材料。Objective To study the physical properties and biological safety of the chitosan-collagen scaffold,and the feasibility of the Chi-Col scaffold as the biomaterial for periodontal tissue engineering.Methods The freeze-drying method was used to prepare mixed scaffolds of Chi-Col in different proportions.The porosity of scaffolds was measured by liquid replacement method.The pore size of scaffolds was observed by scanning electron microscopy,and the safety tests of biological materials,such as intradermal stimulation test and pyogen test,were conducted.Results SEM observation showed that the Chi-Col scaffold had a good porous network structure with a pore size of about 100μm.When the ratio of Chi to Col was 1∶2,the porosity and pore size were most consistent with the demand of tissue engineering materials,with statistically significant difference in comparison with the other groups(P<0.05).Test results of elastic modulus and maximum tensile strength of Chi-Col support materials in different proportions showed that when the ratio of Chi to Col was 1∶2,the difference in elastic modulus and maximum tensile strength was significant compared with those in the other two conditions(P<0.05).The mice were in good condition before and after the acute systemic toxicity test.The intradermal stimulation test on the experimental side was negative,and there was no obvious difference from each control side.Conclusion Chi-Col scaffold is a safe implantation material with good physical properties and biosafety.
