摘 要:为了消除常规反相液相色谱法分析槲皮素时复杂样品中的基质干扰,建立了测定茶叶中槲皮素的柱串联色谱法。柱串联技术同时发挥了两种色谱柱的柱效,改善了基质与目标物的分离问题。考察了不同的色谱柱串联模式对茶叶中槲皮素分离的影响,发现HILIC柱串联C18柱时,槲皮素色谱峰附近的基质干扰可降到最小。在该色谱柱串联模式下,进行色谱条件优化,在最佳分离条件下,测得3种茶叶中槲皮素含量分别为42,48,120μg/g,加标回收率为81.5%~111.9%,RSD为3.0%~8.5%。建立的柱串联反相色谱法可以扩展到其他复杂基质样品的分析。In order to eliminate matrix interference in complex samples for the detection of quercetin by conventional reversed-phase chromatography,a column tandem chromatography method for its determination in tea was established. The column tandem technology simultaneously exerted the column efficiency of two columns,which greatly improved the separation of the target analyte with the matrix. Different column tandem modes were investigated. It was found that the matrix interference could be minimized when HILIC column tandem C18 column mode was used for quercetin analysis in tea sample. The chromatographic conditions were optimized. The contents of quercetin in the three tea samples were 42,48 and 120 μg/g,respectively. The recoveries were81. 5%-111. 9% with the RSD of 3. 0%-8. 5%. The established column tandem reversed-phase chromatography can be extended to the analysis of other complex samples.
