摘　　要：目的探讨磷酸化的P38丝裂原活化蛋白激酶(p-p38-MAPK)、胰岛素受体(INS-R)在人精子中的表达水平与精子活力的相关性。方法抽取2019年7月至2020年6月于安阳市妇幼保健院就诊的因弱精子症而致男性不育的患者共70例,取其精液标本,利用上游法、密度梯度离心加上游法优选精子,分为活力强的上游精子组和活力弱的沉淀精子组,采用逆转录-多聚合酶链反应(RT-PCR)测定INS-R mRNA在精子中的表达情况,采用免疫印迹法测定p-p38-MAPK与INS-R在两组中的相对表达水平,并分析p-p38-MAPK、INS-R与精子活力的相关性。结果 INS-R mRNA在两组精子标本中均有表达,p-p38-MAPK蛋白与INS-R蛋白在两组精子标本中也均有不同程度的表达,但两组INS-R蛋白的表达水平比较差异未见统计学意义(P>0.05),而p-p38-MAPK蛋白在沉淀精子组中的相对表达量(0.43±0.06)高于上游精子组(0.19±0.04),P<0.05。结论 INS-R的表达水平与精子的活力无关,而p-p38-MAPK的高表达可能是精子活力降低的原因之一。Objective To investigate the correlation of the expression of phosphorylated p38 mitogen activated protein kinase(p-p38-MAPK)and insulin receptor(INS-R)in human sperm with sperm motility.Methods A total of 70 male patients with infertility caused by asthenosperm who were treated in Anyang Maternal and Child Health Hospital from July 2019 to June 2020 were selected.The sperm samples of the patients were taken.The sperm samples were selected by upstream method,density gradient centrifugation and upstream optimization method,and were divided into upper sperm group and precipitated sperm group.The expression of INS-R mRNA in sperm was determined by reverse transcription-polymerase chain reaction(RT-PCR).The relative level of p-p38-MAPK and INS-R in the two groups were determined by Western blotting.The correlation between p-p38-MAPK,INS-R and sperm motility was analyzed.Results INS-R mRNA was detected in sperm samples of both groups,p-p38-MAPK protein and INS-R protein was detected in different degrees in the upper sperm group and the precipitated sperm group.There was no significant difference in the expression of INS-R protein between the two groups(P>0.05).While the expression of p-p38-MAPK protein in the precipitated sperm group(0.43±0.06)was significantly higher than that in the upper sperm group(0.19±0.04),P<0.05.Conclusions The expression level of INS-R has no relation to sperm motility,and the high expression of p-p38-MAPK may be one of the reasons for the decrease of sperm motility.
